Objective:Explore PDZ-binding-kinase/T-LAK cell-originated protein kinase(PBK/TOPK)in human breast cancer tissues and their corresponding cancers.The expression in the organization was analyzed and its clinical relevance was analyzed.It provides an important theoretical basis for clinicians to diagnose and prognose breast cancer.Methods:1.120 patients with breast cancer diagnosed in the hospital affiliated to Yan'an University and the pathology department were enrolled.The cancer tissues of each patient and their corresponding adjacent tissues were collected.The collected tissue samples were immersed and fixed with a fixing solution to prepare paraffin tissue sections.The expression of PBK/TOPK protein in experimental tissue samples was detected by immunohistochemical staining.2.Clinical and pathological data of each patient studied in the statistical experiment,including gender,age,maximum diameter of tumor,lymph node metastasis,tumor histological grade of breast cancer,expression of estrogen and progesterone receptor,human epidermal growth factor receptor 2(Her-2)expression,tumor Ki-67 percentage and TNM staging and other related data.3.Correlation analysis between PBK/TOPK protein expression and clinicopathological data of breast cancer patients was performed by SPSS20.0 statistical analysis software.Result :1.Immunohistochemical staining results showed that the expression of PBK/TOPKprotein was detected in the cancer tissues of 78 patients,and the positive expression rate was 65.0%(78/120).The expression of PBK/TOPK protein was detected in the paracancerous group of 31 patients,and the positive expression rate was 25.8%(31/120).The positive rate of PBK/TOPK protein expression in breast cancer tissues was significantly higher than that in adjacent tissues(P<0.05).2.The clinical and pathological data showed that the expression of PBK/TOPK protein in breast cancer was associated with lymph node metastasis(P=0.001<0.05),tumor histological grade(P=0.000<0.05),the expression of Her-2(P=0.002<0.05)and the expression of tumor Ki-67(P=0.008<0.05).And the patient's gender(P=1.000>0.05),age(P=0.374>0.05),tumor maximum diameter(P=0.465>0.05),estrogen receptor(ER)(P=0.136>0.05)and progesterone receptor(PR)(P = 0.184> 0.05)was no significant correlation.3.Further stratification found that in 69 patients with localized early stage(stage I,stage II,and part IIIA(T3 N1 M0))breast cancer,there were 38 cases of PBK/TOPK protein expression in cancer tissues,and the positive expression rate of PBK/TOPK protein was 55.1%(38/69).In 51 patients with locally advanced(except T3N1M0)breast cancer,there were 40 cases of PBK/TOPK protein expression in cancer tissues,and the positive expression rate of PBK/TOPK protein was 78.4%(40/51).PBK/TOPK protein expression was lower in local early stage than in locally advanced breast cancer(P=0.008<0.05).Conclusion:1.The expression of PBK/TOPK protein in breast cancer tissues may be involved in the occurrence,development and metastasis of breast cancer.2.The expression of PBK/TOPK protein in breast cancer may be a marker for predicting the prognosis of breast cancer patients,and may become a new target for tumor therapy.
