Objective To investigate the effects of Prostate Specific Antigen(PSA)on the proliferation,invasion and apoptosis of human castration-resistant prostate cancer(CRPC)22RV1 cells.To provide more evidences for supporting the potential therapeutic target of PSA for CRPC.Methods PSA-targeted shRNA lentivirus vectors containing shPSA1,shPSA2,sh PSA3,sh PSA4 or negative control(NC)were developed,and then were packaged in lentiviral particles,which were then used to infect castration-resistant prostate cancer 22RV1 cells for the generation of sh PSA1-,sh PSA2-,sh PSA3-,sh PSA4-,or NC-22RV1 cells respectively.The expression levels of PSA m RNA and protein were d measured by real-time fluorescent quantitative-PCR(q RT-PCR)and Western blot,respectively.22RV1-derived cells in which PSA was efficiently knocked down were chosen out.The proliferation,invasion abilities and apoptosis of 22RV1 cells were evaluated by CCK-8 assay,Transwell cell invasion assay and flow cytometry respectively.Results The expression levels of PSA m RNA were downregulated in sh PSA1-22RV1,sh PSA2-22RV1,and especially sh PSA3-22RV1 cells which showed the most significantly gene silencing efficacy(P < 0.01).The expression level of protein was significantly lower in sh PSA3-22RV1 cells than in NC-22RV1 cells(P < 0.01).The proliferation abilities of sh PSA2-and sh PSA3-22RV1 cells at 24,48 and 72 h after adherence were obviously inhibited as compared with those of NC-22RV1 cells(all P < 0.01);the invasion ability of sh PSA1-,sh PSA2-and sh PSA3-22RV1 cells were all decreased(P < 0.01).Cell apoptosis analysis by cell cytometry showed that the transfection of sh PSA2 and sh PSA3 construct increased the proportion of apoptotic cells(P < 0.01).Conclusion PSA promotes CRPC 22RV1 cell proliferation,invasion,and inhibites its apoptosis.In addition,the effect of PSA on Pr Ca is dose-dependent.
