| Objective This study was designed to analyze the relationship between different associated symptom of metabolic syndrome(MS)phlegm syndrome(phlegm-yin-vacuity-complex,phlegm-dampcomplex,phlegm-heat-complex)and physicochemical indexes,and to explore the correlation among different associated symptom of MS phlegm syndrome,IGF2BP2 and TCF7L2 genes.It also attempted to reveals the formation mechanism of concurrent syndrome elements of MS phlegm syndrome and provided a reference for clinical diagnosis and treatment of MS.Method 1.Clinical data of MS patients and Non-MS patients were collected to calculate the syndrome elements score by using the method of syndrome elements differentiation.At the same time,clinical blood samples were also collected to test biochemical indicator.According to the patient’s TCM syndrome elements scores,the MS patients were divided into different groups,MS phlegm-yin-vacuity-complex group(phlegm≥100 and yin vacuity ≥100),MS phlegm-damp-complex group(phlegm≥100 and damp≥100),MS phlegm-heat-complex group(phlegm≥100 and heat≥100),at the same time,we also analysis the distribution of syndrome elements.Non-MS patients were concidered as a control group.2.One-way ANOVA and Non-parametric test was used to analyze the correlation between different associated symptom of MS phlegm syndrome and physicochemical indexes.3.TaqMan-MGB probe method was used to detect the expression levels of IGF2BP2 and TCF7L2 genes in blood.Enzyme-linked immunosorbent assay was used to detect the expression of IGF2BP2 and TCF7L2 in serum.One-way ANOVA was used to analyze the correlation among different associated symptom of MS phlegm syndrome,IGF2BP2 and TCF7L2 genes.Result 1.The syndrome elements distribution of different associated symptom of MS phlegm syndrome.(1)The syndrome elements of disease location appearing in MS phlegm-yin-vacuity-complex group with frequency >5% from high to low were: lung,liver,kidney,heart.The syndrome elements of nature of disease appearing in MS phlegm-yin-vacuity-complex group with frequency >5% from high to low were: phlegm,yin-vacuity,qi-stagnation.(2)The syndrome elements of disease location appearing in MS phlegm-damp-complex group with frequency >5% from high to low were: lung,spleen,kidney.The syndrome elements of nature of disease appearing in MS phlegm-damp-complex group with frequency >5% from high to low were: phlegm,damp,qi-stagnation,qi-deficiency,yang-deficiency,cold.(3)The syndrome elements of disease location appearing in MS phlegm-heat-complex group with frequency >5% from high to low were: lung,liver.The syndrome elements of nature of disease appearing in MS phlegm-heat-complex group with frequency >5% from high to low were: phlegm,heat,qi-stagnation.2.The correlation between different associated symptom of MS phlegm syndrome and physicochemical indexes.(1)General conditions: Age: MS phlegm-yin-vacuity-complex group was higher than control group and MS phlegm-damp-complex group,the difference was statistically significant(P<0.05).Body weight: MS phlegm-damp-complex group and MS phlegm-heat-complex group were higher than control group,the difference was statistically significant(P<0.05).BMI: MS phlegm-yin-vacuity-complex group,MS phlegm-damp-complex group and MS phlegm-heat-complex group were higher than control group,the difference was statistically significant(P<0.05),MS phlegm-damp-complex group was significantly higher than MS phlegm-yin-vacuity-complex group and MS phlegm-heat-complex group,the difference was statistically significant(P<0.05).AC: MS phlegm-damp-complex group was higher than control group,MS phlegm-yin-vacuity-complex group and MS phlegm-heat-complex group,the difference was statistically significant(P<0.01).SBP and DBP: MS phlegm-yin-vacuitycomplex group,MS phlegm-damp-complex group and MS phlegm-heat-complex group were higher than control group,the difference was statistical significance(P <0.01).(2)Biochemical indicator: ALT: MS phlegm-yin-vacuity-complex group,MS phlegm-dampcomplex group and MS phlegm-heat-complex group were higher than control group,the difference was statistically significant(P<0.05).AST/ALT: phlegm-yin-vacuity-complex group and MS phlegm-damp-complex group were lower than control group,the difference was statistically significant(P<0.05).GLU: MS phlegm-yin-vacuity-complex group was higher than control group,MS phlegm-damp-complex group and MS phlegm-heat-complex group,the difference was statistically significant(P<0.01).BUN: MS phlegm-yin-vacuitycomplex group and MS phlegm-heat-complex group were higher than MS phlegm-dampcomplex group,the difference was statistically significant(P<0.05).CRE: MS phlegm-heatcomplex group was higher than control group and MS phlegm-damp-complex group,the difference was statistically significant(P<0.05).LDL-C and UA: Although the differences among groups were statistically significant,the adjusted significance P value was not less than 0.05.3.The correlation between different associated symptom of MS phlegm syndrome and IGF2BP2 genes.The expression level of IGF2BP2 gene in blood: MS phlegm-heat-complex group was significantly higher than control group and MS phlegm-yin-vacuity complex group(P<0.05).There was no significant difference on IGF2BP2 protein concentration in serum among these four groups(P>0.05).4.The correlation between different associated symptom of MS phlegm syndrome and TCF7L2 genes.The expression level of TCF7L2 gene in blood: MS phlegm-damp complex group was significantly higher than control group,MS phlegm-yin vacuity complex group and MS phlegm-heat complex group(P<0.05).There was no significant difference on TCF7L2 protein concentration in serum among these four groups(P>0.05).Conclusion 1.The main concurrent syndrome elements of disease location of MS phlegm-yin-vacuitycomplex group and MS phlegm-heat-complex group were: lung,liver,kidney.The main concurrent syndrome elements of nature of disease of MS phlegm-yin-vacuity-complex group and MS phlegm-heat-complex group were: qi-stagnation.MS phlegm-damp-complex group`s main concurrent syndrome elements of disease location were: lung,spleen.MS phlegm-damp-complex group`s main concurrent syndrome elements of nature of disease were: qi stagnation,qi-deficiency,yang-deficiency,cold.2.There was a correlation between different concurrent syndrome elements of MS phlegm syndrome and physicochemical indexes.The increase of age,GLU and UA was related to the formation of MS phlegm-heat-complex.The increase of BMI and AC was related to the formation of MS phlegm-damp-complex.The increase of BUN and CRE was related to the formation of MS phlegm-heat-complex.3.There was a correlation among different associated symptom of MS phlegm syndrome,IGF2BP2 and TCF7L2 genes.The formation of MS phlegm-heat-complex was related to the increase of IGF2BP2 gene mRNA expression.The formation of MS phlegm-damp-complex was related to the increase of TCF7L2 gene mRNA expression. |
