Objective: To observe the effect of Liuwei Dihuang Pill on the apoptotic rate of nucleus pulposus cells and the expression of apoptosis-related factor PrxII in degenerative rabbit intervertebral disc model,and to treat Liuwei Dihuang Pill in the treatment of degenerative disc lumbar pain.The curative effect is supported by experimental evidence,and the path of apoptosis affecting the nucleus pulp osus cells of the intervertebral disc is further explored,which helps to refine the disease caused by degenerative intervertebral disc.Method: According to the random principle,80 New Zealand white rabbits were divided into four groups,namely Liuwei Dihuang Pill Group(Group A),Model Group(Group B),Sham Operation Group(Group C)and Normal Control Group(Group D).Group and Liuwei Dihuang Pills group were used to construct an animal model of intervertebral disc degeneration.After white acupuncture,the skin was disinfected.The skin was cut through the anterior and posterior later al approach of the left abdomen.The fascial muscles were separated according to the level.Care should be taken not to hurt the intervertebral disc.The blood vessels are directed to the anterior side of the vertebral body,and the interverte bral space of L4~L5 and L5~L6 is exposed,and then any one of L4~L5 an d L5~L6 intervertebral discs is randomly selected in the front side of the inter vertebral disc side using a micro syringe with a volume of 10 ?l.Inject 1 ?l of 10 ng/?l TNF-? solution into the annulus fibrosus,and use the rongeur to bite off the transverse process of a vertebral body on the intervertebral disc for use as a mark.After the operation,the skin was sutured and returned to the cage for feeding.The sham operation group used the first two groups of surgical methods and the approach to expose the intervertebral space of L4~L5 and L5~L6,but did not do other treatments,sutured in layers,and rubbed complex iodine in the wound to prevent infection.The blank group was not surgically treated.After 3 days,the penicillin was given 4 million u/qd·im.The Liuwei Dihuang Pill group was treated with Liuwei Dihuang Pill Capsule suspension during the feeding process,and the other three groups were compared with nor mal saline.The rabbits were sequentially killed in batches after 2,4,6,and 8 weeks,and 5 rats were sacrificed at each time point,and the intervertebral disc specimens were reserved.The morphology of the cells was observed by light microscopy.The apoptosis rate was detected by TUNEL reagent,and the expres sion of PrxII protein was detected by Western-blot method.Result:Compared with the sham operation group New Zealand white rabbit(gro up C)and model group(group B),the apoptosis rate and PrxII protein expressi on of the model group were higher than the sham operation group and the nor mal control group in the same feeding cycle.Sex(P<0.05),indicating successful construction of the rabbit degenerative disc model.Compared with the Liuwei Dihuang Pill group,the model group was significantly higher than the Liuwei Dihuang Pill group in the comparison model group New Zealand white rabbit(group B)and Liuweidihuang pill group(group A).<0.05),indicating that Liuwei Dihuang Pill can inhibit the formation of Prx II protein and effectively slow the apoptosis process of nucleus pulposus cells.In the model group,the time points of different specimens in New Zeala nd rabbits were compared between the two groups.The apoptotic rate was grad ually increased,and the PrxII protein content was also increased.The difference was significant(P<0.05).It also showed that the rabbit intervertebral disc dege neration model was successfully constructed.The apoptotic rate and PrxII protein content were gradually increased in the Liuwei Dihuang Pill group New Zealand rabbits at different time points after collection.However,compared with the model group,the trend was small,and there were significant differences between the two groups(P<0.05).),indicating that Liuwei Dihuang Pill has therapeutic significance for degenerative interve rtebral disc.Conclusion:Liuwei Dihuang Pill can moderately slow down the apoptosis of nucleus pulposus cells,down-regulate the expression of Prx II protein,and to some extent treat the degeneration of intervertebral disc.
