Objective: To study the role of miR-150 and its target genes C-MYB,C-MYC in the pathogenesis of peripheral blood mononuclear cells in patients with systemic lupus erythematosus(SLE),and related indicators with SLE laboratory The relationship between clinical disease activity and the role of miRNA-150,C-MYB,and C-MYC in SLE,and provide reliable laboratory indicators for the diagnosis and interpretation of SLE.Methods: 94 patients with systemic lupus erythematosus diagnostic criteria were selected,according to the SLE patients with systemic lupus erythematosus disease activity index(SLEDAI)criteria,divided into high activity group 36 cases(SLEDAI ? 15 points),medium activity group 30 cases(SLEDAI ? 10 points)and 30 cases of low activity group(10 points > SLEDAI ? 5 points)and 44 healthy controls were selected as control group,?The expression levels of miR-150,C-MYB and C-MYC were detected by real-time quantitative real-time PCR(qPCR).Statistical analysis of its relationship with SLE disease activity and SLE related test indicators.A receiver operating characteristic curve(ROC curve)was constructed to analyze the value of miRNA-150 in diagnosing SLE.Results: 1.The expression level of miR-150 in SLE patients was significantly lower than that in healthy controls(P<0.05),the activity group was significantly lower than the middle activity group(P<0.05),and the middle activity group was significantly lower than the low activity group(P<0.05).The low activity group was lower than the healthy control group(P<0.05).2.The expression level of C-MYB in SLE patients was significantly higher than that in healthy controls(P<0.05).The high activity group was significantly higher than the middle activity group(P<0.05),and the middle activity group was significantly higher than the low activity group(P< 0.05),there was no significant difference between the low activity group and the healthy control group(P>0.05).3.The expression level of C-MYC in SLE patients was significantly higher than that in healthy controls(P<0.05).The high activity group was significantly higher than the middle activity group(P<0.05),and the middle activity group was significantly higher than the low activity group(P<0.05).),the low activity group was higher than the healthy control group(P<0.05,).4.Correlation analysis between miR-150 and C-MYB showed a negative correlation between the two(r=-0.724,P=0.012);correlation analysis between miR-150 and C-MYC showed a negative correlation between the two(r =-0.529,P = 0.024).Correlation analysis of indicators: miR-150 was negatively correlated with anti-dsDNA antibody and AnuA antibody(r=-0.482,P=0.021;r=-0.725,P=0.035);C-MYB was positively correlated with SLEDAI score(r= 0.439,P=0.018);CMYC was positively correlated with SLEDAI score and ESR(r=0.609,P=0.012;r=0.456,P=0.032).5.The area under the ROC curve(AUC)for miR-150 diagnosis of SLE was 0.825 [95% confidence interval(95% CI)= 0.745 to 0.905].When the miR-150 cut-off value was 0.53,its sensitivity to diagnose SLE was 65.9%,and the specificity was 87.5%.However,when miR-150 was combined with anti-dsDNA antibody and AnuA antibody to diagnose SLE,the area under the curve(AUC)was 0.913 [95% confidence interval(95% CI)=0.872-0.955].When the cutoff value is 0.71,the sensitivity of the diagnosis of SLE is 80.9%,and the specificity is 90.2%.Conclusion: 1.The expression of miR-150 is decreased in patients with SLE,while the expression of C-MYC and C-MYB is increased in SLE patients,suggesting that miR-150,C-MYC and C-MYB may be involved in the pathogenesis of SLE.2.miR-150 anti-dsDNA antibody and AnuA antibody were negatively correlated.ROC curve showed that miR-150 has certain diagnostic value for SLE.The combined diagnosis of the three is more diagnostic.3.C-MYB is positively correlated with SLEDAI score,indicating that C-MYB can reflect the disease activity of SLE to some extent.4.C-MYC is positively correlated with SLEDAI score and ESR,indicating that C-MYC can better indicate disease activity and is expected to become a new activity index.
