Objectives It is a common complication of postoperative local infection. The experiment is to prepare new antibacterial materials with the new nanoparticles and electrospinning. The physicochemical properties, drug release curves,in vitro sterilization Performance, and further in the rat's infection model compared with the control group after the healing of the wound and the body's response to the situation, and the evaluation of the biological material in the application of the safety and feasibility,in order to find a precise Effective material to reduce the incidence of postoperative infection.Methodology(1)Synthesis of loaded silica-loaded mesoporous silica nanoparticles,in vitro determination of the physical and chemical properties of silver nanoparticles loaded with nanoparticles and drug release, and to assess the sterilization of resistant bacteria.(2)The silver nanoparticles loaded with silver nanoparticles were combined with electrospinning, and their physical and chemical properties and drug release were evaluated in vitro. And to evaluate the antibacterial activity of the resistant bacteria.(3)The model of infected animals was established.SD rats were randomly divided into several groups. Each group was treated with material implantation and the wound was wound with PROLENE.(4)postoperative observation of experimental animal wound healing.(5)The animals were sacrificed in batches, and the level of inflammatory cells in the venous blood of the animals was analyzed. The histopathological changes and immunohistochemical analysis were observed under HE staining.(6)Rats were divided into experimental group and control group. The experimental group was implanted with experimental material, periodically collected blood, analyzed the effect of materials on liver and kidney function, and evaluated their biosafety. Results The electrospinning with nanoparticles loaded with silver nanoparticles has a stable and stable drug release capacity and a stable structure. In vitro antibacterial test confirmed that it can effectively inhibit the growth and reproduction of drug-resistant bacteria.Animal experiments show that it can inhibit the growth of bacteria, conducive to wound healing.Analysis of blood routine results and local tissue analysis and immunohistochemical results show that it can effectively inhibit the inflammatory response, has a good anti-bacterial effect of inhibition of inflammation. The biosafety of the antimicrobial material showed that it didnot cause significant damage to the liver and kidney function of the animal. Its long-term impact on the body to be further studied. Conclusions The new antimicrobial material can effectively inhibit the infection of drug-resistant bacteria, contribute to the healing of the wound,is expected to carry out clinical trials,in order to solve the problem of clinical wound infection to provide a new method.
