Globally, obesity has become a global problem affecting human health. Many of these women are obese because of their diet or their genes.Obesity has a serious impact on the health of women and children.Generally speaking, obesity can damage the ovary and oocytes of women.Female obesity can lead to the decline of oocytes quality, abnormal spindle-chromosome complex(SCCs),aneuploidy and epigenetic changes, and significantly increase reactive oxygen species(ROS).Accumulation of species,ROS.According to a large number of scientific research reports, when ROS abnormally accumulates in the body,it can cause great harm to the body.Phycocyanin(PC)is an antioxidant,neuroprotective,anti-inflammatory and free radical scavenging substance isolated from Spirulina platensis.Previously, our group had studied that in the galactose-induced senile mice model, continuous infusion of PC for 30 days could significantly improve the SCCs abnormality of galactose-induced senile mice and reduce the ROS accumulation in cells. In this paper,we will study whether PC can improve the reproductive function of obese female mice by feeding obese female mice with PC for 45 consecutive days, andexplore the mechanism of its function. The results showed that PC could partially improve the disorder of glucose and insulin metabolism caused by obesity, increase the coefficient of ovary organs, decrease catalase(CAT)activity in ovary and decrease the number of atresia follicles in obese women. At the same time,PC can significantly reduce the fragmentation rate of MII stage oocytes, increase the fertilization rate in vitro, and improve the expression level of H3K9me3 in oocytes. In addition,PC can significantly improve the abnormal SCCs,abnormal mitochondrial distribution, increased ROS level and early apoptosis of female oocytes caused by obesity. These results indicate that PC can significantly improve the oocytes quality of obese female mice, and thus further improve the reproductive capacity of obese female mice.In conclusion, this study found that PC with antioxidant effect played an important role in improving the reproductive capacity of obese female mice. After intragastric administration of PC to obese female mice, not only the oocytes quality of mice was improved, but also the reproductive ability of mice was improved. These results provide a new basis for the clinical application of PC in the prevention and treatment of reproductive defects, such as obesity-related oocytes quality and fertility decline in the future.
