Physalis pubescens L.is an important species of the genus Physalis belonging to the family of Solanaceae.It contains a variety of chemical constituents,such as amino acids,alkaloids,polysaccharides,limonin and microelement.Polysaccharides is one of the most important bioactive constituents.Therefore,it is of great significance to reveal the basis of its active substance by studying the extraction,purification and structural characterization of the polysaccharide from P.pubescens L.The crude polysaccharide was firstly extracted with hot water,and then purified through the column of DEAE-52 and Sephadex G-200 to obtain three polysaccharide fractions?PPLPs?:PPL-P1 and PPL-P2 with white flocculent solid,PPL-P3 with light yellow flocculent solid.The fourier transform-infrared?FT-IR?and nuclear magnetic resonance?NMR?spectroscopy,scanning electron microscopy?SEM?and ultraviolet?UV?were used for the characterization of preliminary structural features of PPLPs.And high performance liquid chromatography?HPLC?and high performance gel permeation chromatography?HPGPC?were used for the molecular weight?MW?and chemical composition analysis.PPL-P1 was mainly composed of mannose,rhamnose,glucose,galactose,xylose and arabinose in a molar ratio of 1.00:0.60:1.37:3.92:0.45:0.67,while PPL-P2 and PPL-P3 were composed of mannose,rhamnose,glucose,galactose and arabinose in a molar ratio of1.00:1.08:1.21:3.48:11.03 and 1.00:5.03:1.64:10.78:16.96,respectively.The molecular weight of PPL-P1,PPL-P2 and PPL-P3 were 7.3KDa,12.1 KDa and 229.3 KDa,respectively.In the study of vitro antioxidant activity,the PPLPs exhibited certain antioxidant activities to DPPH and ABTS radical in a dose-dependent pattern at different concentrations.At the highest concentration of 5 mg/mL,36.2%,41.8%,59.4%of DPPH·were quenched using PPL-P1,PPL-P2and PPL-P3,respectively.The inhibition rate on ABTS radical of PPL-P1,PPL-P2 and PPL-P3 was,respectively,23.1%,12.8%and 43.1%at the highest concentration of 5 mg/mL.In addition,the results showed that PPL-P1,PPL-P2 and PPL-P3 had neuroprotective activity in PPLPs to the H2O2-induced SH-SY5Y cell injury.The results of this study provide a new way for the comprehensive utilization of the resources of Physalis pubescens L.and have certain reference for similar studies.It has a very high medical value,medicinal value,therapeutic value and health value.
