Correlation Between Interleukin-23 Expression And Single Nucleotide Polymorphism Of Receptor Gene And Bone-joint Tuberculosis In Guangxi Zhuang Population | Posted on:2019-08-04 | Degree:Master | Type:Thesis | Country:China | Candidate:Z Y Wu | Full Text:PDF | GTID:2404330596955289 | Subject:Surgery | Abstract/Summary: | PDF Full Text Request | The first part Detection and clinical significance of IL-23 level in peripheral blood of patients with bone and joint tuberculosis in GuangxiObjective: To detect the expression of interleukin-23(IL-23)level in peripheral blood of patients with bone and joint tuberculosis in Guangxi and to explore its clinical significance.Methods: One hundred twenty-five patients with bone and joint tuberculosis admitted to our hospital from July,2014 to March,2017 were selected as the observation group.And one hundred fifty healthy subjects were selected as the control group.The levels of serum IL-23 in peripheral blood were measured by enzymelinked immunosorbent assay(ELISA)both group and the levels of plasma C reactive protein(CRP)and erythrocyte sedimentation rate(ESR)were tested by routine method.The difference of levels of IL-23,CRP and ESR were compared between the two groups and analyze the correlation between IL-23 levels and CRP?ESR levels in observation group.Results:Difference of levels of IL-23?CRP and ESR in peripheral blood of both groups was statistically significant(P <0.01).The levels of IL-23,CRP and ESR in the observation group were significantly higher than those in the control group;The linear correlation between the IL-23 and CRP?ESR levels in peripheral blood in the observation group was not exist(P > 0.05).Conclusion:The expression of IL-23 in peripheral blood of patients with bone and joint tuberculosis in Guangxi was significantly increased.IL-23 may play an important role in the pathogenesis of bone and joint tuberculosis.The second part Association between Polymorphisms of the IL-23 R Gene and Susceptibility of Bone and joint tuberculosis in a Chinese Zhuang PopulationObjective: To analyze the relationship between IL-23 R gene polymorphism and susceptibility to bone and joint tuberculosis in Guangxi Zhuang population.Methods: We selected 125 patients with bone and joint tuberculosis and 150 healthy subjects in a case-control comparison.The samples were collected for the gene polymorphism analysis of IL-23R(rs10489628G/A?rs10489629T/C?rs10889675A/C?rs10889677C/T?rs7530511C/A)by polymerase chain reaction-single base extension technique(PCR)and DNA sequencing method.The study was involved in the detection and comparison of the genotypes and alleles in the two groups.The relationship between IL-23 R gene polymorphism and the susceptibility to bone and joint tuberculersis in Guangxi Zhuang Population was discussed.Results:(1)The frequency of C allele of IL-23 R rs10489629 T/C in the bone and joint tuberculosis group was 29.2% which was higher than that in the control group(21.3%).The frequency of A allele of IL-23 R rs10889675 C/A in the bone and joint tuberculosis group was26% which is higher than that in the control group(18%).The frequency of CA genotype of IL-23 R rs10889675 C/A in the bone and joint tuberculosis group was 37.6% which was higher than that in the control group(25.3%).The difference was statistically significant(P<0.05).(2)The frequency of GTCAC haplotype in the bone and joint tuberculosis group was 69.6% which was lower than that in the control group(77.3%).The frequency of ACACC haplotype in the bone and joint tuberculosis group was 26.0% which was higher than that in control group(17.7%).The difference was statistically significant(P <0.05).Conclusion: The gene polymorphism of IL-23 R rs10489629T/C and rs10889675C/A may be related to the susceptibility of bone and joint tuberculosis in Guangxi Zhuang population.There was no correlation between IL-23 R gene polymorphism and serum IL-23 level in patients with osteoarticular tuberculosis in Guangxi Zhuang population. | Keywords/Search Tags: | Bone and joint tuberculosis, IL-23, CRP, ESR, Interleukin-23, Gene polymorphism, Guangxi Zhuang Autonomous Region, Zhuang Nationality, Osteoarthrosis, Single nucleotide polymorphi | PDF Full Text Request | Related items |
