| Objective:To investigate the distribution frequency of interleukin 33(IL-33)gene rs1342326 and rs928413 Single Nucleotide Polymorphism in guangxi Zhuang children,and to explore the correlation between asthma susceptibility and total serum IL-33 level of black Zhuang children in guangxi area.Methods:(1)In a case-control study,75 children with asthma were selected as the case group,including 45 males and 30 females,and 94 children were selected as the control group,including 52 males and 42 females.(2)Genotypes at rs1342326 and rs928413 of IL-33 gene were detected by the SNa Pshot typing method.ELISA was used to detect the serum level of total IL-33.(3)SPSS 20.0 statistical software was used to analyze the data.The measurement data that did not conform to the normal distribution were represented by the median,and the differences between groups were tested by rank sum test.HWE law was used to test the population representativeness of the samples.Chi-square test was used to compare the genotype distribution and allele distribution between guangxi black clothing zhuang case group and healthy group.Case group risk was expressed as odds ratio(OR)and 95%confidence interval(CI).For the above statistical analysis,P < 0.05 was considered statistically significant.Results:(1)IL-33 gene rs1342326 and rs928413 in both the case group and the control group were in line with Hardy-Weinberg equilibrium law;(2)Polymorphism was found at rs1342326 and rs928413 of IL33 gene.The genotype and allele frequency distribution of rs1342326 locus of IL-33 in the case group and the control group were analyzed.The genotype frequency in the case group was AA 68.0%,AC24.0% and CC 2.7%.The allele frequencies of A and C were 85.3% and 13.3%,respectively.In the healthy group,the genotype frequency was 39.4% AA,58.5% AC and 21.2% CC.The allele frequencies were A 64.4% and C 35.6%,respectively.There were significant differences in genotype and allele frequency(P<0.05).Compared with CC,children with AA genotype had A significantly increased risk of developing asthma(χ 2=3.187,P=0.000,OR=7.4,95%CI= 1.107-26.349).The risk of developing asthma was 2.537 times higher for A allele than for C allele(OR=2.537,95%CI=1.156-2.793).There was no significant difference in genotype and allele frequency between guangxi black cloths case group and healthy group(P>0.05).The distribution frequency of rs1342326A/C locus genotype and allele of IL-33 gene was significantly different among guangxi Zhuang children,Han people in southern China,Europeans,Vietnamese and Americans(P < 0.05).There was no significant difference in the distribution frequency of each genotype and allele of rs1342326 in guangxi Zhuang children compared with African people(P > 0.05).The distribution frequency of rs928413A/G locus genotype and allele of IL-33 gene was significantly different among guangxi Zhuang children,Han people in southern China,Europeans,Vietnamese,Americans and Africans(P < 0.05).(4)The level of serum IL-33 in guangxi black clothing zhuang group was higher than that in healthy group,and the difference was statistically significant(P<0.05).Conclusion:(1)The polymorphism of rs1342326 locus of IL-33 gene is related to the susceptibility of asthma in children of Heiyi Zhuang nationality in guangxi area.(2)The polymorphism of rs928413 locus of IL-33 gene is not related to asthma susceptibility of children of Heiyi Zhuang nationality in guangxi area.(3)The allele frequency distribution of rs1342326 locus of IL-33 gene showed ethnic differences.(4)The allele frequency distribution of rs928413 locus of IL-33 gene showed ethnic differences.(5)The elevated level of IL-33 is related to the occurrence of asthma in guangxi zhuang children. |
