Objective: In this study,spinal synaptosomes of S-D rats were prepared,and qualitative and quantitative analysis of neurotransmitters in rat spinal synaptosomes was performed.Methods: In this experiment,discontinuous Percoll density gradient centrifugation was used to extract synaptosomes from rat spinal cord,and then the morphology and structure of rat spinal synaptosomes were observed under transmission electron microscope.Neurotransmitters in rat spinal synaptosomes were determined by ultra performance liquid chromatography-mass spectrometry.Results: The rat spinal synaptosomes prepared in this study is a morphologically intact and membrane-structured elliptical particle containing synaptic vesicles,mitochondria,cytoplasm and the like.Rat spinal synaptosomes were detected to contain dopamine(DA),norepinephrine(NE),3-methoxytyramine(3-MT),3-hydroxy-2-aminobenzoic acid(3-HAA),Serotonin(5-HT),high vanillic acid(HVA),r-aminobutyric acid(GABA),3,4-dihydroxyphenylacetic acid(DOPAC),5-hydroxyindoleacetic acid(5-HIAA),color Neurotransmitters such as tyrosine(TRP),kynurenine(KYN),and 3-hydroxykynurenine(3-HK).Conclusion: Rat spinal synaptosomes can be prepared by discontinuous Percoll density gradient centrifugation.The neurotransmitters and their related metabolites in rat spinal cord synaptosomes can be detected by liquid chromatography-mass spectrometry.The body is physiologically active. |
