Studies On Traditional Chinese Medicines Against Asthenopia And Preliminary Preparation Of Hydrogels

In this study,based on drug utilizing regularity of traditional Chinese medicine(TCM)by data mining technology and TCM theory,a novel TCM prescription against asthenopia was proposed.Soluble components and essential oils were simultaneously extracted using volatile oil extraction device.The anti-asthenopia activity was evaluated and composition of essential oils was analyzed based on the idea that volatile constituents can stimulate eye mucous membrane to quickly alleviate visual fatigue.Finally,the total extract was preliminarily prepared into hydrophilic gel,and its basic physiochemical properties were determined.1.Global patents related to TCM in treating and preventing asthenopia were systematically searched in IncoPat platform.The TCM prescriptions in the patents were categorized as either“oral”or“external”ones.Then the commonly used TCM were respectively summarized in accordance with the tropism of taste and the type of herbs.The correlation and network graph of the higher frequently used TCM were analyzed by IBM SPSS Modeler 18.0 The difference between oral prescription and external prescription was compared,and the common compatibility of TCM was concluded.the basic law of TCM for the prevention and treatment of asthenopia was discussed and summarized2.Combining the drug utilizing regularity and the TCM theory,a antiasthenopia prescription was generated.On account of the volatile constituents stimulating eye mucous membrane to quickly alleviate visual fatigue,the anti-asthenopia efficacy of essential oils were evaluated by the trials of small intestine smooth muscle contraction,and the content of LDH and Na~+-K~+-ATPase in ciliary muscle of rabbit asthenopia models.3.The volatile oil was extracted by diethyl ether.The chemical components of the volatile oil were qualitatively analyzed by GC-MS.Menthone and camphoras were selected as the indicators,and their contents in volatile oils was determined by gas chromatography method.4.Based on the preliminary screening of gel matrix,moisturizing agents,penetrating agents,antioxidants and surfactant,the prescription of hydrophilic gel matrix was determined.The drug-loading gel was initially prepared by mixing the gel matrix and the extraction of TCM according to certain proportion,and its basic physiochemical properties were analyzed.
