Effect Of Early Treadmill Training Combined With Ultrashort Wave Treatment On Functional Recovery In Rats With Spinal Cord Injury

Objective: Observe the BBB scores,expression of aquaporin-4(AQP-4)and glial fibrillary acidic protein(GFAP)at the 4th weeks after spinal cord injury in rats receive early treadmill training combined with ultrashort wave therapy,therefore to discuss the efficacy and mechanism of simple and combined treatment on spinal cord injury.Methods: 50 female Sprague-Dawley rats were used to make rats SCI(spinal cord injury)models by modified Allen's method.After successfully modeling,40 rats were randomly divided into five groups: sham operation group(only exposed dura mater);the control group(without any other treatment after SCI);the ultrashort wave group(the Allen's strike was firstly performed,and the low-dose ultrashort wave(11 W)treatment then started 24 hours after the SCI,6 min once a day,5 times / week,until the rats were executed);the treadmill group(Allen's strike was firstly given,and the treadmill training started 72 hours after SCI,training time was limited within 20 minutes,once a day,5 times / week,,until the rats were executed);ultrashort wave + treadmill group,(after Allen's strike,the low-dose ultrashort wave treatment started 24 hours after SCI,and the treadmill training started 72 hours after SCI until the rats were executed).On the 1st day,1st week,2nd week,3rd week and 4th week after operation,the motor functional recovery of hind limbs was evaluated by BBB scoring method.Four weeks after operation,the segments of spinal cord encompassing the injury site were removed and immunohistochemical staining was performed.Compare the integrated optical density values ??of protein to observe the expression of GFAP and AQP-4.Results: 1.BBB score: 2 to 4 weeks after operation,all the treatment groups were significantly higher than the control group,the ultrashort wave + treadmill group and the treadmill group were higher than the control group(P<0.01),and ultrashort wave +treadmill group was higher than the ultrashort wave group(P<0.01).The treadmill group was higher than the ultrashort wave group in the 4th week(P<0.05).2.Immunohistochemistry results: in the 4th week after surgery,the expression of AQP-4and GFAP in the control group and the treatment group was significantly increased compared with the sham operation group,and the expression of AQP-4 and GFAP in the three treatment groups was significantly lower than that in the control group(P <0.01);The expression of AQP-4 and GFAP in the ultrashort wave + treadmill group was lower than that in the ultrashort wave group(P<0.01),and the expression of GFAP in the ultrashort wave + treadmill group was lower than that in the treadmill group(P<0.01).The expression of GFAP in the ultrashort wave group was lower than that in the treadmill group(P<0.05).Conclusion: Both the treadmill training and ultrashort wave therapy have therapeutic effects on the recovery of motor function after spinal cord injury in rats.The therapeutic mechanism may be related to the reduction of AQP-4 and GFAP expression in the injured site,and the combination of the two is more effective.
