Study On The Expression Of Myelin Basic Protein In White Matter Damage Of Septic Rats

Objective:To study the expression changes of myelin basic protein and the damage of myelin sheath after white matter damage in septic rats,provide new ideas for the study of sepsis.Methods:64 healthy Wistar rats(18 days old)weighing 30-40 g were randomly divided into two groups: control group and LPS group.LPS group was intraperitoneally injected with 5 mg/kg LPS to prepare rat sepsis animal model;The group was intraperitoneally injected with an equal volume of physiological saline.All surviving animals were anesthetized at the set time at 2h,4h,6h,and 12 h after administration,and the apex was taken and the brain was opened.Myelin injury was observed by staining with Luxol Fast Blue(LFB).Immunohistochemical staining was used to observe the localization of myelin basic protein(MBP)in white matter.Western blot was used to detect the expression of myelin basic protein in rat white brain at different time points.The concentration of MBP protein in serum was detected by enzyme-linked immunosorbent assay(ELISA).Results:Compared with the control group,the expression of MBP protein in the white matter of LPS group was decreased from 2h to 12 h.The expression of MBP protein in LPS group was decreased compared with the control group at 2h,4h and 6h after LPS infection.Statistical significance(P<0.05);although the expression of MBP protein in LPS group was still decreased,it was not significantly different from the control group,and the difference was not statistically significant(P>0.05).The results of immunohistochemistry showed that the MBP optical density of LPS group decreased at 2h,4h and 6h after LPS infection,and the difference was statistically significant(P<0.05).The MBP optical density value of LPS group at 12 h was compared with the control group.Although the comparison was still low,the difference was not significant,and the difference was not statistically significant(P>0.05).The serum ELISA results showed that the serum MBP protein concentration in the experimental group increased at 2h,4h,6h and 12 h after LPS infection,and the difference was statistically significant(P<0.05).Conclusion:In sepsis model rats,MBP protein expression decreased,serum MBP protein concentration increased,indicating that white matter damage caused by white matter damage in sepsis rats caused changes in myelin basic protein and myelin damage.
