| Objective: To investigate the relationship between 163 A/G polymorphism of small ubiquitin-like modified protein 4(SUM04)gene(rs237025)and diabetic retinopathy in northeast China.Methods: 77 hospitalized patients in Chinese Hans with type 2 diabetic retinopathy in our department and 47 healthy Chinese Hans in our physical examination center are selected in June-December,2018.All participants underwent blood sugar,glycated hemoglobin(HbA1c),blood fat,blood serum creatinine,blood urea,uric acid,urine protein,course of disease,disease stage of diabetic retinopathy,high blood pressure level,diastolic pressure and systolic pressure are also took into consideration.SUMO4 163 A/G gene polymorphism is tested by PCR-RFLP.All the data were analyzed by SPSS 25.0 software to test whether the frequency distribution of SUMO4 genotype conforms to Hardy-Weinberg law of genetic equilibrium.Among the measurement data,independent sample t test and one-way analysis of variance(homogeneity of variance should be tested)were applied.Genotype frequency,allele frequency and count data of each group were tested by chi-square test,P<0.05 was statistically significant,and P<0.01 was statistically significant.All blood samples were tested by fixed instruments and analytical methods to avoid measurement errors.Results: The frequencies of AA,GA,GG and GA+GG genotype in the DR group were53.25% 、40.26% 、6.49% 、46.75%.The frequencies of AA,GA,GG and GA+GG genotype in the non-DR group were 65.96%、29.79%、4.26%、34.04%.The frequency of allele A and G in the DR group was 73.38%及 26.62%.The frequency of allele A and G in the non-DR group was 80.85% and 19.15%.There was no significant statistic difference in genotype frequency(P=0.377)and allele frequency(P=0.180)between DR and non-DR groups.There was no significant statistic difference in genotype frequency(P=0.164)of AA and GA+GG between DR and non-DR groups.There was siginificant differences between DR and non-DR groups in blood sugar,glycated hemoglobin,blood serum creatinine,blood urea and uric acid(all P<0.01).There were no statistic differences in blood sugar,glycated hemoglobin,blood fat,blood serumcreatinine,blood urea,uric acid,urine protein,course of disease,disease stage of diabetic retinopathy,high blood pressure level,diastolic pressure and systolic pressure between different genotype(AA,GA and GG)of DR group(all P>0.05).Conclusion: In Han patients with type 2 diabetes mellitus in northeast China,163 A/G polymorphism of SUMO4 gene is not correlated with diabetic retinopathy.No risk factors of diabetic retinopathy were found to be related to the 163 A/G polymorphism of SUMO4 gene in the Han population in northeast China. |
