Objective:To study and investigate the effects and mechanisms of Tetramethylpyra zine(TMP)treated on the cell function(proliferation,apoptosis,metastasis,invasion)of human vulvar squamous cell carcinoma SW962 cells.Methods:Different concentrations culture medium of TMP was used to treat the v ulvar squamous cell carcinoma SW962 cells.The effects of gradient concentration and treatment-time on the proliferation and survival rate of SW962 cells were de tected by MTT assay.The effects of gradient concentration of TMP on apoptosis of SW962 cells was detected by flow cytometry.The effects of gradient concentra tion of TMP on metastasis and invasion of SW962 cells was detected by Transwe ll chamber test and scratch test.Western blot analysis to detect the protein changes of PTEN,AKT,p-AKT and cyclinD1 expression of TMP.Results: TMP significantly inhibited the proliferation of SW962 cells(P<0.05),in duced apoptosis of SW962 cells(P<0.05),increased apoptosis-rate,and inhibited S W962 cell metastasis and invasion(P<0.05).Western blot analysis showed that T MP treatment increased the expression of PTEN and decreased the expression of p-AKT and cyclinD1 protein(P<0.05).Conclusion: TMP can inhibit the proliferation of vulvar squamous cell carcinoma cell line SW962 by inducing PTEN/AKT/cyclinD1 signaling pathway,induce apopto sis and inhibit metastasis and invasion of SW962 cells.
