| Osteoporosis is a common bone degenerative disease in middle-aged and elderly people.It has a high incidence and seriously affects the physical and mental health of patients.At present,most anti-osteoporosis drugs have serious side effects or poor efficacy.Traditional Chinese medicine(TCM)has the characteristics of low price,low toxicity and low side effects,so it has unique advantages in the treatment of such chronic degenerative diseases as osteoporosis.Howerever,traditional Chinese medicine prescriptions have many problems,such as various types of medicinal materials,miscellaneous ingredients,difficult quality control,unclear function mechanism,etc.,which seriously hinder the development and utilization of anti-osteoporosis Chinese medicine products.In this study,we constructed the anti-osteoporosis effective Chinese medicine prescription library to analyze the clinical drug rule of anti-osteoporosis components.Core combinations were extracted by cluster analysis and common TCM combinations were analyzed by correlation analysis.Finally,based on the network pharmacology theoretically research mechanism of the core combination and common combination of traditional Chinese medicine,the MG63 cell model were further used to study and verify the effect and initial mechanism of common anti-osteoporosis traditional Chinese medicine.The main results are as follows:(1)The prescription database of effective traditional Chinese medicine for anti-osteoporosis was constructed,and the clinical medication law of the prescription was analyzed.It was found that most of the anti-osteoporosis prescriptions were tonic drugs,the frequency of warming was the most,Guijing mostly belongs to liver,kidney and large intestine in Chinese medicine.Further,9 groups of anti-osteoporosis core combinations were extracted by KMeans cluster analysis.The types and quantities of Chinese herbal medicines in the nine groups were different.Through correlation analysis of the drug combination rules of the anti-osteoporosis Chinese medicine group,it was found that Epimedium-Rhizoma Drynariae has the highest frequency.(2)The mechanism of the above 9 groups of clusters was analyzed by network pharmacology research.It was found that although the types,quantities and medicinal properties of the traditional Chinese medicines contained in the 9 groups of clusters were different.However,each cluster recipe has an effect on the osteoporosis related signal pathway.We screened 15 anti-osteoporosis differential targets of Epimedium and Rhizoma Drynariae,and analyzed the mechanism of Epimedium and Drynaria by network pharmacology.Further more,the anti-osteoporosis signaling pathways of Epimedium and Rhizoma Drynariae were analyzed.It is found that there are some overlaps and differences in their action pathways.It shows that Epimedium and Rhizoma Drynariae play a role in anti-osteoporosis by acting on different signaling pathways.(3)We studied the compatibility effect of Epimedium-Rhizoma Drynariae with the MG63 cell model,verified the signaling pathways that act,and initially explored possible mechanisms of action.The results showed that Epimedium-Rhizoma Drynariae had a proliferative effect on osteoblasts and significantly promoted the osteogenic differentiation of MG63.In addition,we found that extracts of Epimedium-Bonegrass combination significantly promoted the expression of COL1A1,CAT,IL1 A,IL1B,GSR and RUNX2 genes in MG63 cells,and their effects on the expression of differential proteins similar to the effect of Epimedium extracts and the effect is more pronounced than that of single-flavored drugs. |
