Objective:On the basis of collecting the prescriptions of Traditional Chinese Medicine experts for treating male diseases,this paper discusses the research on the treatment law of male diseases by Chinese medicine,providing the basis for high-quality drug use for optimizing the Traditional Chinese Medicine treatment plan for different types of male diseases and scientific evaluation of effectiveness and safety of clinical application of Traditional Chinese Medicine.Methods:This study collected prescriptions of national famous Chinese medicine experts about four kinds of male diseases(Impotence,Semen Abnormalities,Prostatitis,Benign Prostatic Hyperplasia),based on the "Chinese Medicine Inheritance Auxiliary System(V2.5)"software build a database,and the data mining platform based on Apriori association rules and complex system entropy clustering is used to carry out research to construct a multi-dimensional disciplinary clinical medication data("drug frequency","combination frequency","combination correlation","drug pair correlation coefficient","different core combinations","different new prescriptions")and network display.At the same time,four kinds of male diseases and the highest frequency drugs obtained for them were used for secondary data mining to explore the law of drug use.In addition,on the basis of data mining,the three drugs with higher use frequency were selected as "Cuscutae Semen","Lycii Fructus"and "Epimedii Folium",relying on the Chinese Medicine System Pharmacology Analysis Platform(TCMSP),and then through the construction of Cuscutae Semen pharmaceutical compound-predicted target network,Lycii Fructus pharmaceutical compound-predicted target network,Epimedii Folium pharmaceutical compound-predicted target network,male disease-related PPI network,Cuscutae Semen pharmaceutical compound associated with male disease-potential target network,and Lycii Fructus pharmaceutical compounds-potential target networks,Epimedii Folium pharmaceutical compounds associated with male diseases-potential target networks,potential targets-pathway maps of treatment of male disease with Lycii Fructus,KEGG pathway-target map of treatment of male disease with Epimedii Folium,the action mechanism of Cuscutae Semen,Lycii Fructus and Epimedii Folium is systematically analyzed in treating male diseases.Results:(1)776 prescriptions for the treatment of Impotence were collected.Among them,the top three in the frequency of use are Cuscutae Semen(257 times),Rehmanniae Radix Praeparata(238 times),and Angelica Sinensis Radix(215 times),and the combination of drugs with highest frequency is " Lycii Fructus&Cuscutae Semen "(107 times),"Cistanche Herba&Cuscutae Semen(104 times)," Lycii Fructus&Epimedii Folium"(88 times).The association rules with a confidence level of 0.6 or higher include "Poria->Cuscutae Semen","Rehmanniae Radix Praeparata->Lycii Fructus ","Bupleuri Radix->Angelica Sinensis Radix" and so on.According to the drug association rules and entropy cluster analysis,52 pairs of 3-5 flavor drug core combinations and 26 new prescriptions were evolved.In the treatment of impotence,the highest frequency drug of Cuscutae Semen was in a total of 257 prescriptions,and according to the secondary data mining,the top three use frequency included the Cuscutae Semen(135 times),Cistanche Herba(115 times),Epimedii Folium(100 times).The highest frequency of the combination of the drugs used is"Achyranthis Bidentatae Radix,Cuscutae Semen"(59 times),"Achyranthis Bidentatae Radix,Cistanche Herba"(50 times),"Rehmanniae Radix Praeparata,Epimedii Folium(44 times).Association rules with a confidence level of 0.7 or higher include "Achyranthis Bidentatae Radix->Cuscutae Semen ","Achyranthis Bidentatae Radix->Cistanche Herba".According to the drug association rules and entropy cluster analysis,46 pairs of 3-5 flavor drug core combinations and 23 new prescriptions were evolved.(2)Collected and finished 835 prescriptions for the treatment of Abnormal Semen diseases.Among them,the top three use frequency are Lycii Fructus(442 times),Cuscutae Semen(379 times),and Rehmanniae Radix Praeparata(378 times),and the highest frequency of the combination is " Lycii Fructus,Cuscutae Semen"(295 times),"Rehmanniae Radix Praeparata,Lycii Fructus"(254 times),"Rehmanniae Radix Praeparata,Cuscutae Semen "(223 times).Association rules with a confidence level of 0.9 or higher include"Lycii Fructus,Rubi Fructus->Cuscutae Semen","Rubi Fructus->Cuscutae Semen ".According to the drug association rules and entropy cluster analysis,24 pairs of 3-5 flavor drug core combinations and 12 new prescriptions were evolved.In the treatment of abnormal semen disease,Lycii Fructus is used in a total of 442 prescriptions with the highest frequency drug use,according to secondary data mining,the drugs with top three use frequency were Lycii Fructus(442 times),Cuscutae Semen(295 times),and Rehmanniae Radix Praeparata(254 times),and the highest frequency of the combination of drugs is "Lycii Fructus,Cuscutae Semen "(295 times),"Rehmanniae Radix Praeparata,Lycii Fructus "(254 times),"Lycii Fructus,Epimedii Folium"(199 times).The association rules with a confidence level of 0.9 or higher include "Corni Fructus->Lycii Fructus","Dioscoreae Rhizoma->Lycii Fructus ".According to the drug association rules and entropy cluster analysis,24 pairs of 3-5 flavor drug core combinations and 12 new prescriptions were evolved.(3)Collected and prepared 812 prescriptions for treating Prostate Inflammation.Among them,the top three in terms of frequency of use are Glycyrrhizae Radix Et Rhizoma(354 times),Poria(280 times),and Phellodendri Chinensis Cortex(265 times),and the combination of highest frequency is "Glycyrrhizae Radix Et Rhizoma,Poria"(154 times),"Alismatis Rhizoma,Poria "(123 times),"Plantaginis Semen,Glycyrrhizae Radix Et Rhizoma "(119 times).Association rules with a confidence level of 0.7 or higher include "Anemarrhenae Rhizoma->Phellodendri Chinensis Cortex","Paeoniae Radix Alba->Glycyrrhizae Radix Et Rhizoma " and so on.According to the drug association rules and entropy cluster analysis,52 pairs of 3-5 flavor drug core combinations and 26 new prescriptions were evolved.In the treatment of prostatic inflammation,the drug of Glycyrrhizae Radix Et Rhizoma with the highest frequency has a total of 354 prescriptions,and according to secondary data mining,the first three drugs in terms of use frequency are Glycyrrhizae Radix Et Rhizoma(354 times),Poria(154 times),Plantaginis Semen(119 times);the combination of the drugs used of the highest frequency are "Glycyrrhizae Radix Et Rhizoma,Poria "(154 times)," Plantaginis Semen,Glycyrrhizae Radix Et Rhizoma"(119 times),"Glycyrrhizae Radix Et Rhizoma,Angelica Sinensis Radix"(105 times).The association rules with a confidence level of 0.9 or higher include "Plantaginis Semen->Glycyrrhizae Radix Et Rhizoma","Alismatis Rhizoma->Glycyrrhizae Radix Et Rhizoma"and the like.According to the drug association rules and entropy cluster analysis,28 pairs of 3-5 flavor drug core combinations and 14 new prescriptions were evolved.(4)Collected and prepared 824 prescriptions for the treatment of Benign Prostatic Hyperplasia.Among them,the top three in terms of the frequency of use are Poria(331 times),Achyranthis Bidentatae Radix(320 times),and Astragali Radix(274 times),and the combination of drugs of the highest frequency is "Alismatis Rhizoma,Poria"(172 times),"Achyranthis Bidentatae Radix,Poria "(123 times),"Achyranthis Bidentatae Radix,Persicae Semen"(120 times).Association rules with a confidence level of 0.8 or higher include"Anemarrhenae Rhizoma->Phellodendri Chinensis Cortex","Moutan Cortex->Poria"and so on.According to the drug association rules and entropy cluster analysis,34 pairs of 3-5 flavor drug core combinations and 17 new prescriptions were evolved.In the treatment of benign prostatic hyperplasia,a total of 331 prescriptions associated with the highest frequency drug of Poria;according to secondary data mining,the top three rankings in terms of frequency of use were Poria(331 times),Alismatis Rhizoma(176 times),and Achyranthis Bidentatae Radix(131 times),and the combination of drugs used of the highest frequency is "Alismatis Rhizoma,Poria"(172 times),"Achyranthis Bidentatae Radix,Poria"(123 times)," Glycyrrhizae Radix Et Rhizoma,Poria "(119 times).The association rules with a confidence level of 0.95 or higher include"Corni Fructus->Poria","Rehmanniae Radix Praeparata->Poria" and so on.According to the drug association rules and entropy cluster analysis,46 pairs of 3-5 flavor drug core combinations and 23 new prescriptions were evolved.(5)Analyze and study the multi-component and multi-target complex network of medicine of Cuscutae Semen.The results of the study preliminarily concluded that the active constituents of the Cuscutae Semen drug are playing a role through the targets of PGR,AR,TNF,AHR,CASP8,GSTM1,and IL6.The multi-component and multi-target complex network relationship of Lycii Fructus was analyzed.The results of the study preliminarily concluded that the active components of Lycii Fructus work through targets of PGR,ADRA1A,HTR2A.The multi-component and multi-target complex network relationship of Epimedii Folium drug was analyzed.The results of the study preliminarily concluded that the active constituents of Epimedii Folium drugs work through the targets of AR,ESR1,PGR,SLC6A4,and TNF.Conclusion:(1)Chinese medicine treatment of Impotence is more to use drugs that invigorate the kidney and strengthen Yang,invigorate spleen and benefit sperm,nourish liver and calm heart.Through entropy clustering analysis,new prescriptions such as "Epimedii Folium-Curculiginis Rhizoma-Rehmanniae Radix Praeparata-Lycii Fructus"and "Lycii Fructus-Cuscutae Semen-Rubi Fructus-Rehmanniae Radix Praeparata-Dendrobii Caulis"have the effect of tonifying kidney and strengthening Yang,and according to data mining,the prescriptions of drug Cuscutae Semen of the highest frequency are mostly have the functions of nourishing kidney and strengthening yang,promoting blood circulation and collaterals.For example,the new prescription " Cinnamomi Cortex-Atractylodis Macrocephalae Rhizoma-Morindae Officinalis Radix-Rehmanniae Radix Praeparata-Lycii Fructus-Corni Fructus",which has a good degree of agreement in treating Impotence and has important practical value for clinical drug use experience.(2)Traditional Chinese Medicine treatment of Semen Abnormal diseases mostly use drugs that nourish yin and yang,nourish kidney and nourish blood,and nourish kidney and generate sperms.Through entropy clustering analysis,new prescriptions such as "Schisandrae Chinensis Fructus-Plantaginis Semen-Rubi Fructus-Alpiniae Oxyphyllae Fructus-Dioscoreae Spongiosae Rhizoma","Moutan Cortex-Phellodendri Chinensis Cortex-Polygoni Multiflori Radix-Dried human placenta-Polygonati Rhizoma-Astragali Complanati Semen"all have efficiency of nourishing yin and yang.According to data mining,the drug of medlar prescription of high-frequency in terms of use mostly also have the effect of nourishing yin and yang,replenishing yin and filling the sperm,promoting blood circulation and collaterals and other functions.For example,the new prescription "Eucommiae Cortex-swim bladder-Tougucao-Platycladi Semen-Testudinis Carapacis Et Plastri Colla"has a good agreement with the data of the treatment of Abnormal Semen disease,and has important practical value for clinical medication experience.(3)For Chinese medicine treatment of Prostatic Inflammation,drugs that nourish spleen and kidney,clear heat and dampness,increase urine are used.Through the entropy clustering analysis,the new prescriptions such as "Alpiniae Oxyphyllae Fructus-Cuscutae Semen-Dioscoreae Rhizoma-Alismatis Rhizoma-Moutan Cortex-Poria-Comi Fructus" and "amber-Polygoni Avicularis Herba-Dianthi Herba-Pyrrosiae Folium" have the effect of increasing urine;according to data mining,the highest use frequency drug of Glycyrrhizae Radix Et Rhizoma prescriptions mostly also have the effects of nourishing the spleen and kidney,clearing away heat and dampness,and increasing urine.For example,the new prescription"Coicis Semen-Aconiti Lateralis Radix Praeparat-patriniae-Vaccariae Semen-Salviae miltiorrhizae Radix Et Rhizoma-Manis Squama" has a good agreement with the data of treatment of Prostate Inflammation,and has important practical value for clinical medication experience.(4)Chinese medicine treatment of Benign Prostatic Hyperplasia often use drugs that warm meridians,benefits urine,strengthen liver and kidney,promote blood circulation and phlegm.Through entropy cluster analysis,the new prescriptions such as "Atractylodis Macrocephalae Rhizoma-Polyporus-Alismatis Rhizoma-Poria-Aconiti Lateralis Radix Praeparat-Corni Fructus-Dioscoreae Rhizoma","Akebiae Caulis-Talcum-Dioscoreae Rhizoma-Glycyrrhizae Radix Et Rhizoma-Gardeniae Fructus" have effect of benefiting urine,tonifying liver and kidney.Through data mining,the prescriptions of Fuling with highest frequency of use mostly also have the effect of nourishing yin and increasing urine,benefiting yang and urine,clearing Jin and remove Yuan.For example,the new prescription of "Polyporus-Moutan Cortex-Dioscoreae Rhizoma-Rehmanniae Radix Praeparata-Corni Fructus" has a good agreement with the data of treatment of Benign Prostatic Hyperplasia,and has important practical value for clinical medication experience.(5)Using the network pharmacology method,the multi-component and multi-target complex network relationship of drugs of Cuscutae Semen,Lycii Fructus and Epimedii Folium in the treatment of male diseases were studied.The results of the study preliminarily infer the related mechanisms of Cuscutae Semen,Lycii Fructus and Epimedii Folium in treatment of male diseases,and laid a good foundation for further exploration of its mechanism of action.
