With the rapid development of nanotechnology,the study on nanoscale imaging of sample surface is becoming mature.At present,the nanoscale imaging of sample subsurface structures is one of the hot topics,especially in the field of biomedicaine.The subsurface structures of cells contains abundant information,so it is of great significance in the research area.Acoustic atomic force microscope has both high resolution and three-dimensional detection capabilities.Thus,the application of acoustic atomic force nanoscopic imaging methods in the study of cancer cells can provide help for early diagnosis of cancer,drug screening and evaluation of efficacy.This thesis mainly studies the acoustic atomic force nanoscopic imaging of cancer cells.Firstly,the influence on the cantilever of the contact force between the tip and the sample was analyzed.The simulation model of the cantilever was established the proportional and integral control was included.The imaging parameters were optimized and the images were obtained with good quality.Secondly,the acoustic atomic force nanoscopic imaging technology method of cancer cells was studied.The effects of different curing agents,time durations and concentrations on the imaging of cancer cells were discussed.The conclusion is that the best morphologyical image condition for the lung adenocarcinoma cell was cured by 4% concentration of paraformaldehyde for 60 min and the best acoustic image condition for the same cell was cured by 4% concentration of paraformaldehyde for 10 min.The images of lung adenocarcinoma cell A549 and liver cancer cell SMMC-7721 were compared,and the acoustic images could reflect the subsurface ultrastructure differences of the two kinds of cells.Finally,the application of acoustic atomic force nanoscopic imaging method for cancer cells was studied.The imaging of the cancer cells that treated with the anticancer drug fullerenol and magnetic nanoparticles was studied,respectively.Through the control experiments and acoustic images,the "disappearance" of the ultrastructure of cancer cells on the surface and subsurface was confirmed to be associated with the fullerenol action.By analyzing the acoustic image and the elastic modulus of the cell,the position detection of magnetic nanoparticles in the cells was implemented.The application of acoustic atomic force nanoscopic imaging technology in the study of cancer cells can provide an effective way to observe the ultrastructure of the cell subsurfaces and the distribution of the magnetic nanomaterials.This approach with nanoscopic imaging technology is useful for cancer detection and treatment.
