Walnut?Juglans regia?is the fruit of Juglans mandshurica,a deciduous tree of the family Juglandaceae.The wood diaphragm in the core of walnut is called Diaphragma Juglandis Fructus,which Uygur people always use it to bubble water for tea,having the effect of sleep and tonifying the kidney.The cyan outer pericarp of unripe walnut is called Green walnut husks,which has a long history of folk medicine.At present,the researches on Diaphragma Juglandis Fructus and Green walnut husks are still not thorough enough,so that the inland people often do not recognize the value of them,resulting in environmental pollution and waste of resources.In order to develop and utilize the resources reasonably and explore the possibility of using Diaphragma Juglandis Fructus and Green walnut husks into medicine,this experiment studied the chemical constituents of them and provided the basis for the further development of the medicinal value of them.The research's main contents are as fellows:1.In this experiment,Diaphragma Juglandis Fructus and Green walnut husks were respectively separated and purified by silica gel column chromatography,reversed-phase C18 column chromatography,MCI medium pressure chromatography,LH-20 gel column chromatography and preparative HPLC,the structures were determined by spectroscopic methods and spectral technique.Sixteen compounds were isolated from Diaphragma Juglandis Fructus,including?-?-syringaresinol?1?,?+?-pinoresinol?2?,?+?-?7R,7'R,7''S,7'''S,8S,8'S,8''S,8'''S?-4'',4'''-dihydroxy-3,3',3'',3''',5,5'-hexamethoxy-7,9';7',9-diepoxy-4,8'';4',8'''-bisoxy-8,8'-dineolignan-7'',7''',9'',9'''-tet raol?3?,2,3-dihydroxy-1-?4'-hydroxy-3'-methoxy-phenyl?-propan-1-one?4?,3-hydroxy-1-?4'-hydroxy-3'-methoxyphenyl?propan-1-one?5?,3',4'-dimethoxy phenylpropanediol?6?,?2S?-3,3-di-?4-hydroxy-3-methoxyphenyl?-propane-1,2-diol?7?,3-hydroxy-1-?4'-hydroxy-3',5'-dimethoxyphenyl?propan-1-one?8?,?1R,5R,6R?-6-{4'-O-[8''-?7''-?4''-hydroxyl-3''-methoxypheny??glyceol]-3',5'-dimethoxyphenyl}-3,7-dioxabicyclo[3.3.0]octan-2-one?9?,curcasinlignan B?10?,evofolin-B?11?,?7S,8R?-dihydrodehydrodiconifer lalcohol?12?,pinnatifidanin C I?13?,?+?-?7S,8S?-4,1'-dihydroxy-3,3',5'-trimethoxy-7,8,9-trinor-8,4'-oxyneolignan-7,9-diol?14?,dysosmarol?15?,and1-?4'-hydroxy-3'-methoxyphenyl?-2-[4''-?3-hydroxy propyl?-2'',6''-dimethoxyphenoxy]propane-1,3-diol?16?.Fourteen compounds were isolated from Green walnut husks,including 5-hydroxy-4-methoxy-1-tetralone?17?,4,8-dihydroxy-1-tetralone?18?,4,5-dihydroxy-?-tetralone?19?,blumenol B?20?,dehydrovomifoliol?21?,megastigm-5-ene-3,9-diol?22?,juglanin B?23?,blumenol C?24?,loliolide?25?,oleracone B?26?,methyl 4-hydroxy-3-methoxybenzoate?27?,isovanillic acid?28?,and?-?-syringaresinol,?+?-pinoresinol which have been isolated from Diaphragma Juglandis Fructus.2.The anti-tumor activity of compound 1-28 was tested by MTT method in vitro.Cervical cancer cell?Hela?,human gastric cancer cell?HGC-27?and colon cancer cell line?Ht-29?were used as target cell lines.The results showed that compounds 4,6,7,12,18,19,23,25 and 27 showed different inhibitory activities on these three kinds of tumor cells in vitro,respectively.Among them,compound 23 showed the strongest inhibitory activity against three kinds of cancer cell lines,followed by compounds 6and 18 showed good inhibitory activity on Hela cells and Ht-29 cells.In this experiment,the chemical constituents of Diaphragma Juglandis Fructus and Green walnut husks were studied systematically.It was found that there were abundant chemical constituents in both of them,among which the lignans were the most in Diaphragma Juglandis Fructus.Then the antitumor activity of the isolated compounds was screened in vitro.The results showed that both of them contained compounds of antitumor activity in vitro,suggesting that both of them have the potential to develop new anticancer drugs.
