Objectives:After transplantation of human umbilical cord mesenchymal stem cells(HUC-MSCs)into MRL/lpr lupus mouse model,whether HUC-MSCs can reduce the immune response of SLE mice,miRNA181a and osteopontin expression levels in kidney tissues.Methods:Twenty-four 22-week-old female MRL/lpr mice were randomly divided into experimental group and control group.0.5 ml of 1×106 HUC-MSCs were infused into the experimental group MRL/lpr mice through the tail vein,and the same volume of normal saline were infused into the control group MRL/lpr mice through the tail vein.1.Observe the body weight and 24-hour urine protein changes of experimental and control MRL/lpr mice HUC-MSCs before and after treatment.2.Enzyme-linked immunosorbent assay(ELISA)was used to detect ANA and anti-double-stranded DNA(ds-DNA)antibody titer levels in two groups of MRL/lpr lupus mice.Immunohistochemistry and Western blotting were used to detect the expression of OPN in kidney tissue of MRL/lpr lupus mouse model before and after stem cell treatment.4.RT-qPCR was used to observe the expression of miRNA181a in kidney tissue of MRL/lpr mouse model before and after stem cell treatment.Results:After 3 weeks of treatment,the body weight of the experimental group(22.73±0.31)g was higher than that of the control group(21.43±0.15)g(P<0.05),and the urine protein of the experimental group(0.39±0.02)mg was significantly lower than the control group(0.71±0.04).The difference was statistically significant(P<0.05).2.After 3 weeks of treatment,the anti-ds-DNA and ANA antibody titers in the experimental group were lower than those in the control group(P<0.05).The expression of OPN in the kidney of MRL/lpr lupus mice treated with HUC-MSC transplantation was significantly lower(P<0.05).The expression of miRNA181a in the kidney tissue of MRL/lpr lupus mice treated with HUC-MSC transplantation was significantly lower(P<0.05).Conclusions:The transplantation of HUC-MSC delayed the weight loss of MRL/lpr lupus mice,the urinary protein content increased.The transplantation of HUC-MSC decreased ANA and anti-ds-DNA antibody titers in serum of MRL/lpr lupus mice.The expression of OPN and miRNA-181a in the kidney tissue of MRL/lpr lupus mice were reduced,and the symptoms of renal injury were alleviated.
