Objectives:1.To investigate the relationship between the serum neuregulin-1(NRG-1)level and the gamma activity of the prefrontal cortex of electroencephalogram(EEG)in the resting state with clinical symptomsin first-episode schizophrenia patients characterized,as well as cognitive dysfunction.2.To futher study the changes of NRG-1 and the gamma activity and the correlation of them in patients with schizophrenia. Methods:The serum levels of NRG-1 in 53 patients and 58 controls were measured by enzyme-linked immunosorbent assay(ELISA).The gamma activity were collected from the lead of FP1 and FP2 of the prefrontal cortex of EEG.The patients' psychotic symptoms were assessed by positive and negative syndrome scale(PANSS).The MATRICS consensus congtive battery(MCCB)was used to assess the cognitive function. The differences of the serum NRG-1 levels, the gamma activity and the sub-score of MCCB between the two groups were compared. The correlation between the serum level of NRG-l,as well as the gamma activity and the sub-score of MCCB was analyzed in the two groups. And the correlation between the serum level and the gamma activity was analyzed in the two groups. The correlation between the serum level,as well as the gamma activity, and the score of PANSS were analyzed in the case group. Results:?The serum levels of NRG-1 in the case group was lower than that in the control group, the difference was statistically significant[(7.36 ± 3.96)pg/mL vs(11.02± 8.78)pg/mL,P<0.01];The gamma activity was significantly different between the case group and the control group(?2=27.140,P=0.000).?There was a negative correlation between the serum NRG-1 level and the gamma activity in the case group(r=0.542,P<0.01),while the correlation of them is no significance in the control grouP.?There was a negative correlation between the serum NRG-1 level with PANSS total score(r=-0.360,P<0.05),while the gamma activity was positively correlated with PANSS total score(r=0.278,P<0.05).?The scores of TMT in MCCB in the patients was higher than that of the control group(P<0.01),while the scores of BACS SC,HVLT-R,NAB,BVMT-R,and CF scores were lower than those of the control group(P<0.01).?The serum NRG-1 level was significantly positively correlated with the scores of HVLT-R in the case group(r=0.332,P<0.05),and the gamma activity was significantly negatively correlated with the scores of HVLT-R(r=-0.442,P<0.01)and NAB(r=-0.307,P<0.05).Conclusions:?The serum level of NRG-1 in patients with with first-episode schizophrenia was lower than that in healthy controls and negatively correlated with PANSS total score. The percentage of gamma activity in EEG in the cases group was significantly higher than that in healthy controls and positively correlated with PANSS total score. These suggest that patients with schizophrenia have abnormal neurobiochemical and neurophysiological responses in the early stage, and the degree of symptoms may be related to the lack of neurotrophic factor and the level of neurotransmission, which provides new potential biomarkers for early identification, diagnosis and treatment of schizophrenia.?The serum NRG-1 level in the patient group was negatively correlated with the gamma activity of EEG,indicating that the pathogenesis of schizophrenia in neurobiochemistry and neuroelectrophysiology is not independent of each other, and may be an inhibitory relationship.?The serum NRG-1 level in the case group was positively correlated with the HVLT-R score in the MCCB cognitive test. The gamma activity of the EEG was negatively correlated with the HVLT-R and NAB scores respectively. These suggest that neurobiochemistry and neuroelectrophysiology may have a certain relationship with cognitive dysfunction of schizophrenia. At the same time, the serum NRG-1 level and gamma activity are related to the HVLT-R score, which suggests that both may participate in some pathological mechanisms related to cognitive dysfunction.
