Objectives:(1)The levels of bacterial lipopolysaccharide(LPS)and its toll-like receptor 4(TLR4)in serum of patients with liver cirrhosis were measured.(2)Analysis of changes in the intestinal flora of patients with cirrhosis.(3)To explore the correlation between LPS and TLR4 expression in the peripheral blood of patients with liver cirrhosis,and provide a new perspective for the detection of liver cirrhosis and clinical prevention.Methods:(1)Determination of LPS concentration in peripheral blood of patients with liver cirrhosis by spectrophotometry.(2)The level of TLR4 in peripheral blood of patients with liver cirrhosis was measured by Elisa method.(3)Descriptive analysis of the published literature on the changes of intestinal flora in patients with cirrhosis by means of historical literature review.Results:(1)Among the 18 samples,there were 3 cases with CTP grade A,LPS concentration<10pg/ml,9 cases with CTP grade B,and LPS concentration<10 pg/ml to 48.14 pg/ml.There were 6 cases with CTP grade C,and the LPS concentration ranged from 105.6 pg/ml to 1000 pg/ml.Spearman's grade correlation analysis was used in the three groups.The difference was statistically significant(p=0.0001).<0.001).(2)The corresponding TLR4 level of samples with elevated LPS levels also increased significantly,and the difference was statistically significant(p<0.05).(3)20 articles were included in the changes of intestinal flora in patients with cirrhosis,and 18 of them showed an increase in Gram-negative bacteria.Conclusions:A review of the literature on intestinal flora analysis in patients with(1)The level of LPS in serum of patients with cirrhosis is significantly up-regulated,and its concentration is positively correlated with CTP grade of liver function.(2)The level of serum TLR4 in patients with cirrhosis showed an upward trend and was positively correlated with LPS concentration.(3)There is a disorder of intestinal flora in patients with cirrhosis,and the number of Gram-negative bacteria is mainly increased.cirrhosis and a significant up-regulation of LPS and TLR4 levels in the experimental results indicate that there is a significant intestinal flora imbalance in patients with cirrhosis.And LPS into the blood further aggravates liver damage,affecting the development of the disease.Therefore,LPS and TLR4 assays are expected to become new indicators of cirrhosis assessment,and intestinal flora may be a new target for the prevention and treatment of cirrhosis.
