| Objective:A rapid and sensitive ultraperformance liquid chromatography coupled with mass spectrometry(UPLC-Q-TOF/MS)method has been developed for simultaneous determination of 58 abused drugs and their metabolites(Pholcodine,Morphine,Hydromorphone,Cathinone,Codeine,Ephedrine,Methcathinone,Methylephedrine,Caffein e,Mescaline,03-monoacetylmorphine,O6-monoacetylmorphine,Amphetamine,MD A,Hy drocodone,MA,MDMA,PMMA,Benzoylecgonine,MDEA,Nokethamide,Ketamine,6-acet ylcodeine,Thebaine,Heroin,Cocaine,Dihydroetorphine,7-aminoflunitrazepam,Pethidine,Z olpidem,LSD,Phenacetine,Fenfluramine,Phencyclidine,Chlordiazepoxide,Clozapine,Bro mazepam,Midazolam,Flurazepam,a-Hydroxymidazolam,Buprenorphine,Zaleplon,EDDP,a-Hydroxyalprazo lam,a-Hydroxytriazolam,Nitrazepam,Oxazepam,Estazolam,Clonazepa m,Lorazepam,Methadone,Methaqualone,Alprozolam,Triazolam,Flunitrazepam,Temazepa m,Diazepam,Prazepam)in urine.Methods:The samples were determined by UPLC-Q-TOF/MS method after processed with dilution method,protein deposition method.The target compounds were separated by Acquity UPLC HSS C18 column and gradient eluted with 0.1%formic acid-acetonitrile and 5 mmol/L ammonium formate-water(pH=3)as the mobile phase.The electrospray ionization in positive ion was used for analysis in full scanning mode(MSE).UNIF1 software was used to establish the database and to process samples.Results:The extraction effect of the methanol precipitation is the best for 58 abused drugs after comparing with the extraction recoveries and the matrix effect of three sample processing methods for human urine samples.The 58 target compounds were all detected by the established database in UNIFI software.Conclusion(s):This paper has established an UPLC-Q-TOF/MS method to detect 58 abused drugs and their metabolites in human urine samples,the method presented here is validated to be simple,rapid,sensitive and accurate for the determination of trace medicines and metabolites in urine samples,which could be used for the pretreatments in court on a scientific laboratory analysis of 58 abused drugs and their metabolites in human urine samples. |
