Objective:The objective of this study was to evaluate the effects of naringin on the growth,acid production,sugar production and adhesion of major cariogenic bacteria and biofilms in vitro,and to explore its potential anti-caries mechanisms.Methods:1.Determining the minimum inhibitory concentration?MIC?,the minimum bactericidal concentration?MBC?,the minimum biofilm inhibition concentration(MBIC50),the minimum biofilm reduction concentration(MBRC50)and drawing growth curve of naringin on Streptococcus mutans,Streptococcus sanguis,Streptococcus sobrinus,Actinomyces viscosus,Actinomyces naeslundii and Lactobacillus acidophilus by liquid dilution experiment.To evaluate the effects of naringin on the planktonic state and biofilm growth of 6 species of cariogenic bacteria;2.Based on the changes in pH difference??pH?of six major cariogenic bacteria and biofilms,naringin was used to determine the effect of naringin on the bacterial planktonic state and the biofilm acid production capacity,and a dynamic acid production curve was drawn to evaluate the effect of bacterial acid production;3.Determining the effect of naringin on the ability to synthesize water insoluble extracellular polysaccharides?IEPS?by three major sugar-producing bacteria,Streptococcus mutans,Streptococcus sanguis and Actinomyces viscosus;4.Three strains of Streptococcus mutans,Streptococcus sanguis and Actinomyces viscosus were selected as experimental strains to determine the effect of naringin on the ability of the three bacteria to adhere to inclined test tubes.Results:1.The MICs of naringin on Streptococcus mutans,Streptococcus sanguis,Streptococcus sobrinus,Actinomyces viscosus,Actinomyces naeslundii and Lactobacillus acidophilus were 1,0.5,0.5,0.5,1 and 1 g/L;MBCs were 2,1,1,1,2 and 2 g/L;MBIC50were 2,2,4,1,2 and 2 g/L;MBRC50 were 4,4,8,4,4 and 4 g/L.Naringin can inhibit the growth of planktonic state and biofilms of the major cariogenic bacteria;2.Naringin can inhibit the acid production process under the planktonic state and biofilms of the six cariogenic bacteria;3.Naringin can inhibit the synthesis of IEPS at MIC,1/2 MIC concentration of Streptococcus mutans,and MIC concentration of Streptococcus sanguis,which was statistically significant?P<0.05?;4.Naringin can inhibit the adhesion process of Streptococcus mutans and Actinomyces viscosus at the concentrations of MIC and the following 4 levels,showing a dose-dependent trend,and the difference between each experimental group and the control group was statistically significant?P<0.001?.Conclusions:Naringin can inhibit the growth,acid production,sugar production,and adhesion of Streptococcus mutans,Streptococcus sanguis,Streptococcus sobrinus,Actinomyces viscosus,Actinomyces naeslundii and Lactobacillus acidophilus in the planktonic state,and has shown effectively inhibited and cleared the biofilm and inhibited the acid production process of the biofilm,and understood the pharmacological effects and potential mechanisms of naringin on caries prevention.
