Study On Pharmacodynamics And Toxicology Of Zicaosanhuang Suppository

Objective: To compare and confirm the efficacy and safety of Zicaosanhuang suppository in the treatment of hemorrhoids,and lay the foundation for its clinical application and further research and development of new drugs.Methods:1)Established animal models of inflammation and measured the number of inflammatory factors at the inflammatory site,studied the anti-inflammatory effect of the drug;The animal pain models were established and the content of pain factors were measured to investigate the analgesic effect of the drug;Established the models of bleeding and congestion in animals and measured the clotting index to explore the hemostasis of the drug;The bacteriostatic effect of the drug was verified by the test of bacteriostatic intensity and potency?the content and electrical conductivity of the bacterial cell contents.2)Acute toxicity was observed and recorded within 2 weeks after rectal administration of drugs to mice;3)Rats in different dose groups were given rectal administration for 1 month,and the long-term toxicity was observed after 1 month of rectal administration and 2 weeks after drug withdrawal,the hematology?blood biochemical indexes and organ coefficients of each group were compared and the pathological examination was performed.Results: 1)Compared with the positive drugs,there were no significant differences between the high dose group and the middle dose group in most of the pharmacodynamic experiments and the effects of regulating related factors(P>0.05).2)Acute toxicity experiment mice were alive and healthy after being administered 776 times the daily clinical dosage of adults,and the body weight and organ coefficient of the administration group were not statistically different from those of the blank group(P>0.05).3)In the long-term toxicity experiment,there were no abnormality in the whole cycle of each dose group.Compared with the blank group,there were no statistical difference in blood cytology,blood biochemistry and the coefficient of organs(P>0.05).Pathological sections showed no significant pathological changes in major organs.Conclusion: The anti-inflammatory,analgesic,hemostatic and bacteriostatic effects of the Zicaosanhuang suppository are superior to or similar to the positive drugs.At the same time,the preparation has high safety and is suitable for clinical treatment of hemorrhoids.
