Objective:The basal study is that researched the protective effect of Ziziphus jujuba Mill ethanol and water extract on the multiple organs damage in the HAPH model rats.Methods:(1)50 SD rats were randomly divided into five groups,which were plain control group(CG),plateau model group(MG),nifedipine group(NE),Ziziphus jujuba Mill ethanol extract group(ZJM-AG)and Ziziphus jujuba Mill water extract group(ZJM-WG),and 10 rats in each group.10 rats were fed under plain conditions for 45 days,40 rats were fed under plateau condition of 5 000 m altitude for 30 days.Based on the successful establishment of the rat model of high altitude pulmonary hypertension,40 SD rats were randomly divided into four groups.The five groups were measured pulmonary arterial pressure(PAP).Serological indexes of rats were detected by enzyme-linked immunosorbent assay,such as c-reactive protein(CRP),erythropoietin(EPO),vascular endothelial growth factor(VEGF),angiotensinogen(AGT),apolipoprotein E(Apo-E)and apolipoprotein AⅠ(Apo-AⅠ).(2)To observe the morphological changes of pulmonary artery,lung,heart,brain,liver and kidney tissues of rats in each group.(3)The serum of rats were analyzed by ~1H-NMR metabolomics.Results:(1)Compared with the CG,the PAP of the MG increased,with statistical significance(P<0.05);Compared with the MG,the PAP of the NE,ZJM-AG and ZJM-WG decreased,with statistical significance(P<0.05).Compared with the CG,the serological indexes of CRP,EPO,VEGF and AGT in rats respectively rise in the experiment of MG,NE,ZJM-AG and ZJM-WG,the serological indexes of Apo-AⅠand Apo-E decreases steadily,with statistical significance(P<0.05);Compared with MG,the study found that CRP,EPO,AGT,VEGF in rats decrease in the experiment of NE,ZJM-AG and ZJM-WG,the serological indexes of Apo-AⅠand Apo-E rises,with statistical significance(P<0.05).(2)Under light microscope,Ziziphus jujuba Mill extract can improve the morphology of pulmonary artery,lung,heart,brain,liver and kidney tissues of rats in the HAPH model rats.(3)A total number of 19 different metabolites were identified by ~1H-NMR metabolomics,including amino acids(glutamine,taurine),saccharides(glucose),lipid(VLDL),and so on.Conclusion:Ziziphus jujuba Mill extract has therapeutic effect on HAPH model rats,which the mechanism may be via improved the level of serological indexes,adjusted the amino acids metabolism,saccharometabolism and lipid metabolism,decreased pulmonary artery pressure,indirectly improved metabolic homeostasis reaction,enhanced the oxidation resistance of the body.Hence,Ziziphus jujuba Mill extract significantly improved the injury of multiple organs in HAPH model rats.
