Objective: In this study,we screened circRNA and mRNA differentially expressed between blood samples from patients with tuberculosis and control populations and verified large samples in the population to determine relevant biomarkers.We also screened and verified miRNAs in order to explain the molecular level and Mechanisms of TB infection.Methods: The whole blood samples of 3 patients with pulmonary tuberculosis and 3 patients in the control group were collected and sequenced by the transcriptome sequencing to obtain the expression profile of circRNA and mRNA.In order to preliminarily determined the target gene.We collect 83 cases of tuberculosis patients and 90 healthy controls in Xinjiang who are the same region,gender and age.At the same time,blood samples of 43 patients with other lung diseases including pneumonia and lung cancer were collected for RT-qPCR verification to analyze the expression of target gene.The miRNA chip was then used to correlate the differential circRNA with mRNA results in order to find genes and pathways related to tuberculosis infection.Results: 1)Whole transcriptome sequencing results showed 835 differentially expressed circRNA in the blood samples of pulmonary tuberculosis patients in Xinjiang,of which 249 were up-regulated and 586 were down regulated.There were 1218 differential mRNAs,including 430 up-regulated and 788 down regulated;besides,there were 76 up-regulated miRNAs and 92 down-regulated miRNAs 2)The difference mRNA was enriched by go and pathway analyse;3)The results of RT-qPCR showed that the down-regulation of the expression of hsa-circ-0008276 and hsa-circ-0003452?P<0.05?was consistent with the results of microarray;4)The results were combined with the target miRNA of different circRNA,and hsa-miR-574-5p and hsa-miR-574-5p were selected for further analysis.Conclusion: 1)The difference of circRNA between the tuberculosis group and the healthy control group was screened out by using the whole transcriptome chip,which provided the thinking for the development of tuberculosis;2)hsacirc0003452,hsacirc0008276 have a certain correlation with tuberculosis,of which hsacirc0008276 has a high correlation with tuberculosis,and the area under the ROC curve is 0.7296,indicating that it can be used as a potential marker of tuberculosis;3)has-circ-0008276 may play a important role in the occurrence and development of tuberculosis by competing with the target gene has-miR-574-5p.
