Study On The Relationship Between Klotho Gene And Idiopathic Calcium Oxalate Renal Calculi In Uygur And Han Nationality In Xinjiang

Objective: To investigate the association between rs3752472,rs564481 and rs145682430 polymorphism of Klotho gene and idiopathic calcium oxalate nephrolithiasis in Uygur and Han nationality in Xinjiang.Methods: 399 Uygur ICOS patients(case group),410 Uygur healthy subjects(normal group),412 Han ICOS patients(case group)and 400 healthy Han patients(normal group)were selected.PCR method and DNA sequencing were used to determine the distribution frequency of rs3752472,rs564481,rs145682430 genotypes and alleles of Klotho gene polymorphisms,and to analyze the correlation between related gene loci and idiopathic calcium oxalate nephrolithiasis.Results: the distribution of genotypes in the selected Uygur,Han and normal groups were consistent with Hardy-Weinberg balance,and the study population was representative.The C allele of rs3752472 was the protective factor of ICOS in Han nationality(P<0.05,OR=0.498,95%CI: 0.332~0.734),CC genotype was 0.518 times higher than that of T genotype(95%CI: 0.349~0.768,P<0.05),and the C allele of rs3752472 was different between Uygur case group and Han case group(P<0.05,OR=0.566,95%CI:0.394~0.814),rs564481 allele C was different between Uygur normal group and Han normal group(OR=0.553,95%CI:0.441~0.693,P=0.000);there was difference between Uygur case group and Han case group(OR=0.597<1,95%CI=0.477~0.751,P=0.000);and rs145682430 allele C was significantly different between Han case group and Uygur case group(OR=5.507,95%CI:2.292~13.21,P=0.000).Conclusion: The C allele of Klotho gene rs3752472 locus is the protective factor of ICOS in Xinjiang Han population,while rs145682430 and rs564481 allele C is more likely to appear in Han population(case group and normal group),but there is no significant correlation with Han and Uygur population.
