Objective:This study was based on bio-impedance technology to determine the reference value of intracellular and extracellular body fluid relatedindexes of normal rats.Based on this,the change situations of intracellular and extracellular body fluid related indexes of normal rats after intervention and pathological state rats were studied.Methods:?1?150 rats,male and female in equal,use Bio-Imp Vet Body to masure Total Body Water/Body Weight?TBW/BW?,extracellular fluid/total body water content?ECF/TBW?,intracellular fluid/total body water content?ICF/TBW?,fat mass/body weight?FM/BW?,free fat mass/body weight?FFM/BW?,extracellular fluid/intracellular fluid?ECF/ICF?in rats.?2?196 rats were weighted and evenly divided into normal group(25g?d-1,30ml?d-1),1.8%Na Cl group,50%glucose solution group,spironolactone group,hydrochlorothiazide group,furosemide group and water restriction group in a total of seven groups,there were 28 rats in each group,male and female in equal,The rats were intervened only one group and measure the indexes of this group every day for six consecutive days,The drinking restriction group restricted drinking water three days in advance.Bioimpedance technology was used to measure the body fluid indexes of rats in each experimental intervention group,At the time points of 0.5,1,2,4,6,8,and 10hours after the intervention,the body fluid indexes of the rats were measured with Bio-Imp Vet Body.After finding the six groups experimental intervention rats of response time point,experimental intervention was performed every day for four weeks in a row;the body fluid indexes of all rats were measured one,two,three,and four weeks after the intervention.?3?Twelve male SD rats,Six rats were randomly selected as normal group,and the remaining six rats were injected with a single tail vein injection of doxorubicin?6 mg/kg?to reproduce a rat model of nephrotic syndrome,which was divided into model group,the body fluid indexes of the normal group and the model group were respectivelymeasured in the fourth week.?4?Twenty male SD rats,tenrats were randomly selected as normal group,and the remaining ten rats were taken combine with Unclean eating combined and overwork to duplicate the model of spleen deficiency and wetness.The body fluid indexes of the normal group and the model group were measured in the fourth is?0.798±0.074?:1.?2?Comparison of reference values of body fluid indexes between male and female rats The results showed that TBW%of male ratswas?51.01±2.87?%,femalewas?47.99±3.39?%,TBW%of male ratswas higher than that of female rats,ECF%and FM%of male was respectively?43.23±3.27?%and?30.06±3.07?%,ECF%and FM%of female was respectively?45.07±4.68?%and?33.49±5.76?%,ECF%and FM%of male rats were lower than that of female rats.?3?Changes in body fluid indexes and organism response time point in rats with different timegradients under the same intervention degreeThe response time points of time and set a longer gradient to observe the situation of changes in body fluid indexesafter intervention,the impedance was temporarilymeasured after 10 hours.?4?Changes of body fluid indexes in rats under different intervention degreesThe body fluid indexes change of rats in each group in the first week.The results showed that compared with the normal group,TBW%in the 1.8%Na Cl group decreased to?47.4±4.1?%,FM%increased to?35.1±5.6?%,TBW%in the 50%glucose group decreased to?48.5±4.0?%,and TBW%in the hydrochlorothiazide group decreased to?49.6±5.8?%The TBW%in the water-restricted group decreased to?47.8±5.6?%,which were significantly different?P<0.05?.The body fluid indexes change of rats in each group in the second week.The results showed that compared with the normal group,the TBW%in each intervention group significantly decreased,and the FM%significantly increased;the ECF%in the 1.8%Na Cl group rose to?45.6±2.7?%,and There are more obvious edema and sodium retention phenomena all in significant differences.The body fluid indexes change of rats in each group in the third week.The results showed that compared with the normal group,TBW%in each experimental group showed a significant decrease,and FM%increased significantly;the ECF%in the 1.8%Na Cl group,the hydrochlorothiazide group,and the water-restricted group all increased significantly,1.8%.The Na Cl group continued to exhibit edema and sodium and sodium retention,both of which had significant differences.With the deepening of the experimental intervention in the fourth week,the changes of body fluid composition in each intervention group became more obvious toward the purpose of intervention,all of which had significant differences.?5?Changes of body fluid indexes in experimental rats with nephrotic syndromeThe results showed that the ECF%content in the model group was?54.20±4.13?%,which was significantly higher than that in the normal group.?6?Changes of body fluid indexes in rats with spleen deficiency and dampnessThe results showed that the TBW%content in the model group was?57.79±6.56?%,which was significantly higher than that in the normal group;the fat content was?21.05±8.98?%,which was significantly lower than that in the normal group.Conclusions:?1?The indexes of intacellular and extracellular body fluid in the normal group were relatively stable,and the TBW%of male rats was higher than that of female rats,and ECF%and FM%were lower than that of female rats.?2?The response time pointsof Spironolactone and hydrochlorothiazide earlier than sodium supplement and osmotic diuretics?50%glucose?and the high-efficiency diuretic furosemide;under experimental intervention,rats will produce negative feedback regulation that weakens this intervention.?3?The body fluid indexes in each intervention group were changed by changing the osmotic pressure and water intake environment.As the degree of experimental intervention continued to deepen,the changes in body fluid indexes became more obvious.?4?Significant edema and water and sodium retention in rats were appeared with nephrotic syndrome.?5?Spleen deficiency and water-dampness internal cease rats showed significant weight loss and water-fluid metabolism disorders.
