Objective:Summarize the research status of antler,compare the chemical components of different antler processed products,and provide a theoretical basis for antler quality control.To study the protective effects of different antler processed products on rats with kidney-yang deficiency.Methods:1.The Kjeldahl method,2,3-dinitrosalicylic acid colorimetry method and biological kit were used to determine the content of total protein,total polysaccharides and sex hormones in antler,and the differences in the chemical composition of different antler processed products were compared;2.Investigate different extraction solvents,different extraction methods,different extraction times and times,and optimize the extraction conditions of antler nucleosides;3.HPLC method using methanol?A?-0.07%acetic acid?B?as mobile phase to determine the content of six nucleosides in antler and conduct methodological investigations.Establish antler using"Chinese medicine chromatographic fingerprint similarity evaluation system"Fingerprint of nucleosides;4.Carry out one test and multiple evaluations of antler nucleoside components,calculate the relative correction factors of the other 5 components with hypoxanthine as the reference peak,and conduct reproducibility surveys;5.Cluster analysis,principal component analysis?PCA?and least squares discriminant analysis?OPLS-DA?using the contents of 6 nucleosides in antler as variables and look for differential compounds;6.Forty male SD rats of 180-220 g SPF grade were selected,and hydrocortisone injection was used to reproduce a rat model of kidney-yang deficiency with intraperitoneal injection at a dose of 20 mg·kg-1.Shenqi Pill 2.08 g/?kg·d?and antler 400 mg/?kg·d?for30 days Index,observe the pathological changes of kidney and testis to evaluate the efficacy of different velvet processed products.Results:1.The average value of total protein content in freeze-dried antler,bloody antler,and blood-extracting antler was 71.49%,64.35%,and 56.45%respectively;the average value of total polysaccharide content was 3.01,1.33,1.64 g·kg-1;The average content was 2.65,1.84,2.26 ng·g-1.The overall analysis showed that the content of active ingredients in freeze-dried antler was higher than that of boiled and fried antler;except for the protein content of blood antler,it was lower than that of blood-extracting antler.2.Optimize the extraction conditions of antler nucleosides:accurately weigh the appropriate amount of antler powder?pass 40 mesh sieve?,add 20 times the volume of distilled water,sonicate for 30 min,centrifuge(9800 r·min-1)for 10 min,and take the supernatant The solution was placed in a 50 mL volumetric flask and filled with water to make up to the mark.3.An HPLC method was established for antler nucleosides,and 6 chromatographic peaks were identified as uracil,adenine,hypoxanthine,uridine,inosine,and guanosine,and antler nucleoside components were established.The fingerprints,the similarity of 30batches of velvet antler were all greater than 0.95.4.The results of one test and multiple evaluations suggest that the relative correction factors for the other five components calculated using hypoxanthine as the reference are0.944,1.172,0.481,0.533,and 0.584,and the reproducibility of the correction factors is examined.The RSD values are less than 2%,indicating that the reproducibility is good,and the measured value of the external standard method is not significantly different from the calculated value of QAMS.5.After modeling,the rats suffered from chills,back deflation,and weight loss,indicating successful modeling.The body weight increased significantly after administration;the levels of serum CORT,ACTH,CRH,TSH,CRE,BUN,and cGMP in the rats after the model were significantly increased,which were significantly different from those in the blank group?P<0.05?.Decreased,there was a significant difference with the blank group?P<0.05?,no significant change in cAMP content?P>0.05?;all indicators were improved after the drug intervention,and the DG group had a better effect.Conclusion:The chemical composition of freeze-dried antler are higher than that of boiled and fried antler.Except for the protein content,the contents of active ingredients of dermatophytes are higher than those of bloody antler.There was no significant difference in blood fungus.
