Obiective:To improve the quality standard of Angelica sinensis and Angelica sinensis pached with alcohol;To analyze the quality evaluation index of Angelica sinensis through literature collation and optimize the processing technology of Angelica sinensis pached with alcohol by single factor test to provide theoretical basis for obtaining the best processing technology of Angelica sinensis pached with alcohol;Using modern analytical techniques,to establish a better quality control method and draft the quality standard of Angelica sinensis pached with alcohol.Methods: 1.Study on the Improvement of Quality Standard of Angelica sinensis Following the requirements and analytical methods of Angelica sinensis in the Chinese Pharmacopoeia of 2015 edition,the characteristics,microscopic identification,TLC identification,moisture,total ash,acid insoluble ash,volatile oil and ferulic acid content of Angelica sinensis were analyzed.In addition,on the basis of Chinese Pharmacopoeia in 2015 edition,the DNA barcode of Angelica sinensis was identified by plant DNA barcode technology,the fingerprint of Angelica sinensis was determined by high performance liquid chromatography,the heavy metal content of Angelica sinensis was determined by inductively coupled plasma mass spectrometry,the ligustilide content of Angelica sinensis was determined by high performance liquid chromatography.2.Study on Processing Technology of Angelica sinensis pached with alcohol The single factor experiment was conducted with the parameters of frying time,frying temperature,moistening time and the amount of rice wine,and with the evaluation indexes of the content of ferulic acid,ligustilide and alcohol extract.To obtain the best processing technology of Angelica sinensis pached with alcohol through the analysis of single factor test results,and carry out the verification of process.3.Study on Quality Standard of Angelica sinensis pached with alcohol Following the requirements of Angelica sinensis pached with alcohol in the Chinese Pharmacopoeia of 2015 edition,the properties,microscopic identification,TLC identification,moisture,total ash,acid-insoluble ash and alcohol extracts of Angelica sinensis pached with alcohol were analyzed,and the content of ferulic acid and ligustilide was determined by high performance liquid chromatography and the content limit was specified.4.Comparative Study on Chemical Constituents of Angelica sinensis before and after Wine-processing By comparing the fingerprints of Angelica sinensis with those of Angelica sinensis pached with alcohol before and after wine processing,to discuss the differences in chemical constituents between Angelica sinensis and Angelica sinensis pached with alcohol.Results: 1.The properties and microscopic identification results of Angelica sinensis were in line with the requirements of Chinese Pharmacopoeia 2015 edition.TLC identification results of Angelica sinensis: fluorescent spots of the same color appeared in the chromatographic positions of the samples corresponding to the chromatographic positions of the control medicines and the control substances.The water content of Angelica sinensis ranged from 4.8% to 12.4%.The total ash content ranges from 3.9% to 8.0%,and acid insoluble ash content ranges from 0.3% to 2.3%.The contents of heavy metals and harmful elements in Angelica sinensis were determined as follows: lead content ranged from 0.17 mg/kg to 0.81 mg/kg,cadmium content ranged from 0.018 mg/kg to 0.690 mg/kg,mercury content ranged from 0.0043mg/kg to 0.0310 mg/kg,copper content ranged from 5.9 mg/kg to 13.1 mg/kg,arsenic content ranged from0.21mg/kg to 0.82mg/kg,and total heavy metals ranged from 6.5 mg/kg to 14.7 mg/kg.The samples were identified as Angelica sinensis(Oliv.)Diels by DNA barcode identification test in the DNA barcode identification system of Chinese medicinal materials.There were 18 common peaks in the fingerprint of Angelica sinensis.The overall similarity of chromatograms of Angelica sinensis in each batch ranged from 0.927 to 0.999.The content of alcohol-soluble extracts of Angelica sinensis in each batch ranged from 45.4% to 61.8%.The content of Volatile oil ranged from 0.3% to 1.2%,The content of ferulic acid ranged from 0.0552% to 0.1366%,The content of ligustilide ranged from 0.4976% to 2.3822%.2.The optimum processing technology of Angelica sinensis pached with alcohol is as follows: take the slices of Angelica sinensis,mix with 15% yellow rice wine,put them in a closed container for 1 hour,and put them in a preheated electric frying pan with a set temperature of 200 ?,fry them for about 15 minutes,take them out and cool them off.3.The Content of 8 batches of Angelica sinensis pached with alcohol,the content of water ranged from 4.26% to 7.49%,total ash ranged from 5.2% to 7.5%,acid insoluble ash ranged from 0.8% to 2.2%,alcohol extract ranged from 50.41% to 65.24%,ferulic acid ranged from 0.0329% to 0.0626%,and ligustilide ranged from 0.4840% to 0.9625%.4.The overall similarity of the chromatograms of Angelica sinensis pached with alcohol in different batches is between 0.796 and 0.995.The overall similarity of the chromatograms of Angelica sinensis in different batches is between 0.926 and 0.998,and the similarity is good.Three common peaks were reduced on the chromatogram of Angelica sinensis pached with alcohol on the basis of chromatogram of Angelica sinensis,and two new common peaks have emerged.The peak area of each common peak changed in varying degrees after wine processing.Conclusion: Through systematic study of Angelica sinensis and Angelica sinensis,the quality standard of Angelica sinensis was drafted and worked out,which is higher than that of Chinese Pharmacopoeia(2015 edition).The processing standard of Angelica sinensis with quantified technology and easy operation was optimized.The quality standard of Angelica sinensis was drafted and worked out,which is higher than that of Chinese Pharmacopoeia(2015 edition).The chemical composition of Angelica sinensis before and after alcoholization would change to some extent.Therefore,it provides a research basis for guaranteeing the quality of Angelica sinensis and wine Angelica sinensis and improving the quality of Xiaojin Capsule.
