Objective:Proteomics method was used to analyze the serum of patients with GA-ONFH by mass spectrometry,explore the key indicators of You Gui Yin intervention in GA-ONFH,reveal the mechanism of You Gui Yin in treating kidney-yang deficiency type GA-ONFH,and provide modern medical evidence for dialectical treatment of GA-ONFH by TCM And provide modern ideas for the prevention and treatment of GA-ONFH.Methods:? Sixty patients with hormone-related femoral head necrosis who underwent hip-preserving surgery in the joint department of the Jiangsu Provincial Hospital of Traditional Chinese Medicine were included.The time span was 2019.03-2020.01.The ARCO stage of the femoral head necrosis was ? a-? a.Deficiency type,peripheral serological specimens were taken as intervention group before operation(preoperative).All patients underwent core decompression+dead bone removal+ bone graft+ fibula rod support.One week after the operation,all patients were treated with oral righteous drink for 60 consecutive days.After the end of the course of treatment,peripheral serological specimens were again taken as the intervention group(postoperative).Mass detection was performed on the above serological samples.? Including 10 volunteer patients with hip osteoarthritis hospitalized due to non-femoral head necrosis as the control group,the peripheral serological specimens of the patients were collected and the same mass spectrometry was performed.? The serum mass spectrometry results of the intervention group(preoperative)and the control group were compared first,and the proteins significantly related to hormone-related femoral head necrosis were initially screened.Then observe the changes of these proteins in the intervention group(after operation)after the intervention of You Gui Yin,so as to find out the proteins related to the intervention of You Gui Yin.? The enrichment analysis of GO functional annotations and KEGG pathway annotations on differential proteins was made to speculate the signaling pathways that Yougui drink may participate in in the intervention process,and to explore the mechanism of Yougui drink in treating kidney-yang deficiency type hormone-related femoral head necrosis.Result:? This study finally screened a total of 9 different proteins such as YAP protein that are significantly related to the effect of You Gui Yin on kidney-yang deficiency hormone-related femoral head necrosis.? Differential protein functions in GO enrichment analysis are mainly related to molecular functions such as transcription factor binding,?-catenin binding,and BH domain binding.Differential proteins in the results of KEGG enrichment analysis are mainly related to four signaling pathways including Hippo signaling pathway.Conclusion:You Gui Yin mainly affects the molecular functions of transcription factor binding,0-catenin binding,and BH domain binding.And mainly intervened the Hippo signaling pathway,Wnt/?-catenin signaling pathway,thyroid hormone signaling pathway,P53 signaling pathway,and right-going drink through the above-mentioned mechanism played a role in the treatment of kidney-yang deficiency hormone-related femoral head necrosis.
