Objective: To study the expression of LncRNA XIST and mi RNA-101 in pancreatic cancer tissues and pancreatic cancer cells and the relationship between them,further study the regulation of LncRNA XIST on miRNA-101 and the effect of LncRNA XIST on the growth and metastasis of pancreatic cancer.Methods: Samples of tumor tissues and adjacent tissues removed from patients with pancreatic cancer after surgical treatment were collected,and two types of pancreatic cancer cells were cultured at the same time.The expressions of LncRNA XIST and miRNA-101 in pancreatic cancer tissues and cells were calculated,and the expression of both Correlation between quantities.Double luciferase experiments were performed to verify the unidirectional regulatory relationship between LncRNA XIST and miRNA-101.Two types of pancreatic cancer cells were transfected.The transfected cells were XIST knockdown group,XIST enhanced group,and For control transfection,Real-time PCR was used to calculate gene expression and verify the accuracy of transfection.After transfection,the cells of each group were tested for cell viability(CCK-8),cell proliferation test(scratch test),migration invasion test(Transwell),and then co-transfection of two genes to obtain Control,shXIST,shXIST + miR-NC,shXIST + miRNA-101 inhibitors(Panc-1 group)and Control,XIST,XIST + miR-NC,XIST + miRNA-101 mimics(BxPc-3 group).Finally,pancreatic cancer cells transfected with XIST-enhanced,XIST-knockdown and control groups were implanted subcutaneously in the groin of nude mice,and the tumor formation volume was measured.To study the effect of LncRNA XIST on the growth and metastasis of pancreatic cancer by inhibiting the expression of miRNA-101.Results: LncRNA XIST expression was higher in pancreatic cancer tissues,and miRNA-101 expression was lower.LncRNA XIST expression was lower in paracancerous tissues,miRNA-101 expression was higher,and miRNA-101 expression and LncRNA XIST was negatively correlated(P <0.05).In the Panc-1 group,the expression of miRNA-101 in each transfected cell gradually increased,and the OD value was significantly different(P <0.05),indicating that the transfection was successful and the corresponding transfection XIST knockout cell scratch test showed that the cell proliferation ability was reduced.Transwell experiments showed that the distribution of cancer cells in the lower compartment was reduced and the invasion ability was reduced(P <0.05);the expression of miRNA-101 in each transfected cell in the BxPc-3 group gradually decreased,and the OD value was significantly different(P <0.05),suggesting that The successful transfection and corresponding XIST-enhanced cell scratching experiments showed that the cell proliferation ability was increased,and the Transwell experiment showed that the number of cancer cells in the lower compartment increased significantly and the invasion ability was enhanced(P <0.05).In nude mice tumor formation experiments,the tumor formation volume of nude mice enhanced by XIST transfection was larger than that of nude mice transfected with XIST(P <0.05).Conclusion: LncRNA XIST can promote the proliferation and invasion of pancreatic cancer cells,and LncRNA XIST can inhibit the antitumor effect of miRNA-101 in pancreatic cancer cells.
