LncRNA-Dicer-AS Inhibits Invasion And Metastasis Of Pancreatic Cancer Through Regulating Dicer Transcription | | Posted on:2021-03-12 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Li | Full Text:PDF | | GTID:2504306104992379 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Purposes This study explored the effect of Dicer-AS on proliferation,invasion and metastasis of pancreatic cancer cells through dicer1,further explored the biological mechanism of the occurrence and development of malignant progression of pancreatic cancer.All of this is to provide a new strategy for the comprehensive treatment of pancreatic cancer and prolong survival time.Methods According to the data sets GSE14245,GSE41368 and GSE16515,lnc RNA Dicer-AS was down-regulated.q RT-PCR was used to demonstrate the expression differences of lnc RNA Dicer-AS in pancreatic cancer cell lines and normal pancreatic duct cells.By transfection of Dicer-AS-si RNA and pc DNA-Dicer-AS,the MTT experiment,transwell experiment and scratch experiment were carried out to observe the effect of Dicer-AS on the proliferation,invasion and metastasis of pancreatic cancer cells.Dicer-AS was up-regulated or down-regulated in the same way to observe the changes of dicer1 in m RNA and protein levels.Then the rescue experiment was carried out with the co-transfected plasmids pc DNA-Dicer-AS and dicer1-si RNA to study whether Dicer-AS causes the morphological changes of pancreatic cancer cells via affecting the expression of dicer1.Results We found that lnc RNA Dicer-AS was down-regulated in data sets GSE14245,GSE41368 and GSE16515.Then q RT-PCR showed that the expression of Dicer-AS was lower in pancreatic cancer cells.Further experiments demonstrated that up-regulation or down-regulation Dicer-AS could inhibit or promote proliferation,invasion and metastasis of pancreatic cancer cells.Moreover,overexpression or knockdown Dicer-AS could increase or decrease the transcription and translation levels of dicer1.In the end,we confirmed that Dicer-AS exerted the antitumor effect through promoting the expression of dicer1.Conclusions Dicer-AS promoted expression of dicer1 to inhibit the malignant behavior of pancreatic cancer cells.Inhibiting the malignant behavior of the tumor by promoting the expression of Dicer-AS may become a biological treatment for pancreatic cancer to prolong the survival time. | | Keywords/Search Tags: | Long non-coding RNA, pancreatic cancer, Dicer-AS, dicer1, invasion, migration, proliferation | PDF Full Text Request | Related items |
