Objective: To explore the mechanism of Astragalus polysaccharide,an effective component of Astragalus membranaceus,in the treatment of allergic rhinitis guinea pig model based on Treg/ Th17 immune balance,and try to expand the research boundary of Th1/Th2 homeostasis,which is common in the pathogenesis of allergic rhinitis.Meanwhile,to explore the effect of Astragalus Polysaccharide on Treg/Th17 related cytokines Foxp3 and ROR?t and the expression level of IL-17 in serum of guinea pig?To explain The mechanism of Astragalus Polysaccharide in the treatment of allergic rhinitis partly.Methods: 60 healthy male guinea pigs were randomly divided into 5 groups: A,B,C,D and E(A astragalus polysaccharide injection group,B astragalus polysaccharide nasal spray group,C glucocorticoid nasal spray group,D allergic rhinitis model group n and E ormal control group).According to the classical modeling method,group A,group B,group C and group D were given basic sensitization: ovalbumin(OVA)0.3mg + Al(OH)30mg + 1ml of normal saline suspension were injected intraperitoneally 7 times a day,local excitation: on the first day after basic sensitization,50 ?L of 2% ova solution on each side was dripped into the nose for 7 days,once a day.Group E,i.e.the normal control group,was injected intraperitoneally with 30 mg Al(OH)and 1 ml normal saline at the same time,and locally stimulated with the same amount of normal saline.After the preparation of the model,the behavioral scores were determined,and the dead and unqualified guinea pigs in each group were eliminated.Group A was given astragalus polysaccharide injection 10 mg / kg once a day for three weeks.In group B,astragalus polysaccharide injection was dissolved in normal saline to prepare the solution,and nasal spray was conducted once a day according to 1mg / kg.Group C was treated with budesonide nasal spray at a dose of 20 ug / kg once a day.In group D and E,the same amount of normal saline was used instead of nasal spray once a day.The expression of Foxp3 and ROR ? T,which are closely related to Treg/Th17 cells,and the expression of IL-17 in serum of guinea pigs were detected by immunohistochemistry and enzymelinked immunosorbent assay.According to the experimental data,the measurement data are expressed by mean ± standard deviation(?x ± s).Spss22.0 and Excel 2019 software are used for statistical analysis.If the independent samples of multiple groups obey the normal distribution,one-way ANOVA is used.SNK test is used for the comparison between the two groups.If they do not fully conform to the normal distribution,Kruskal Wallis test is used H)If the difference between the two paired samples obeys the normal distribution,the paired t-test is used;if the difference does not obey the normal distribution,Wilcoxon signed rank sum test(paired rank sum test)is used;Spearman correlation test is used for correlation analysis;P < 0.05 is statistically significant.Results:(1)the FOXP3 positive cell rate in the nasal mucosa of a astragalus polysaccharide injection group,B astragalus polysaccharide nasal spray group and C glucocorticoid nasal spray group was significantly higher than that of E normal control group,the ROR?t positive cell rate was lower than that of E group,and the serum IL-17 expression concentration was significantly higher than that of E group(P < 0.01),but the difference was statistically significant(P < 0.01),but it was significantly higher than that of D allergic rhinitis model group There was no significant difference in Foxp3,ROR?t and IL-17 between groups A,B and C(P > 0.05).Conclusion: APS can increase the expression of Foxp3,reduce the expression of ROR?t,correct the imbalance of Treg / Th17 cells,and inhibit the production of IL-17,so as to reduce the symptoms of allergic rhinitis and alleviate the inflammatory reaction of the nose.
