Objective: This study detected the levels of autophagy-related proteins in nasal mucosa of AR,as well as cytokines associated with Treg/Th17 cells in peripheral blood.Then measured the expression of autophagy-related proteins in peripheral blood mononuclear cells.To explore the mechanism of autophagy involved in allergic rhinitis by regulating the imbalance of Treg/Th17 cells.Methods:1、30 people with AR were diagnosed with deviated nasal septum or hypertrophic rhinitis were selected as the experimental group,and 30 patients with simple septum deviated without AR were selected as the control group.2、The nasal tissues were collected during the operation.(1)transmission electron microscopy(TEM)was used to observe the quantity of autophagosomes in the nasal mucosa.(2)the other part is used to detect autophagy-related molecules by Western blot.3、Peripheral blood were collected and divided into two parts.(1)In order to determine whether Treg/Th17 cell is imbalanced in AR,Flow cytometry was used to detect the expressions of cytokines associated with Treg/Th17 cells.(2)In order to study the effect of autophagy on Treg/Th17 cell imbalance in AR,Flow cytometry was used to detect the expressions of autophagy-related proteins in Th17 and Treg cells.Result: 1、Transmission electron microscopy results showed that the content of autophagic vesicles in the nasal mucosa tissue of AR patients was significantly higher than that in the control group.2、Western blot was used to detect the expression of autophagy-related proteins in the nasal mucosal tissues.Compared with the control group,the expression of autophagy-related proteins Beclin1,LC3-II/LC3-I in the AR group was increased,and the expression of P62 was reduced.It shows that autophagy regulates the imbalance of Treg/Th17 cells in AR.3、the results of flow cytometry showed that The expression of IL-17 A in PBMC of AR group was higher than that in control group.While the content of Foxp3 in the AR group was significantly lower than that in control group.The expression of autophagy markers Beclin1 and LC3-II/LC3-I in Th17 and Treg cells of AR group was significantly higher than that of control group,and the expression of P62 was lower than that of control group.It is speculated that autophagy regulates the imbalance of Treg/Th17 cells in allergic rhinitis.Conclusion: The study showed that Autophagy may participate in AR by regulating the imbalance of Treg/Th17 cells.
