BackgroundColorectal cancer is the third most common malignancy in the world,and metastasis is the leading cause of death.Tumorigenesis is a multistep,multi-stage,multi-gene complex process.Recent studies have found that PXMP4 is involved in the development of prostate cancer.But the function of PXMP4 in colorectal cancer is unknown.We found that PXMP4(peroxisomal membrane protein 4)is a differentially expressed gene between colorectal cancer tissues and normal colorectal mucosa by bioinformatics analysis.Experimental study on gene transfection,proliferation,invasion,migration in vitro: To investigate the role of PXMP4 in proliferation,invasion,migration of colorectal cancer,and to provide a new target for the diagnosis and treatment of colorectal cancer.ObjectivesSix colorectal cancers of LOVO,RKO,SW620,SW480,HCT116 and DLD1 were used as research objects to analyze the expression of PXMP4,and the effects of PXMP4 on the proliferation,invasion and migration of colorectal cancer cells were studied by cell function experiment.Methods1.Bioinformatics methods such as the gene expression omnibus(GEO)database were used to analyze the expression of PXMP4 in paired colorectal cancer and normal tissues.2.Immunohistochemistry(IHC)was used to detect the expression of PXMP4 in paraffin samples of colorectal cancer,and the correlation between the expression of PXMP4 and the clinicopathological parameters such as tumor size,differentiation and lymph node metastasis was analyzed.3.The expression of PXMP4 mRNA and protein in six colorectal cancer cells was detected by qRT-PCR and western blot.4.Colorectal cancer cell lines that overexpressed and interfered with PXMP4 were constructed and the expression of PXMP4 was verified by qRT-PCR and western blot.5.Transwell,CCK-8 and scratch assay were used to detect the effects of PXMP4 on the proliferation,invasion and migration of colorectal cancer cell lines.Results1.Gene expression omnibus(GEO)database shows that PXMP4 is a differentially expressed gene in colorectal cancer tissues and normal tissues.2.IHC results showed that PXMP4 was mainly located in the organelle membrane and showed a brownish yellow color.In 112 cases of colorectal cancer patients,the expression of PXMP4 in 80 cases of colorectal cancer tissues was higher than that in the paired normal mucosal tissues.The expression of PXMP4 had no difference with the age,sex and tumor size of the patients(P>0.05),but was closely related to the degree of differentiation and lymph node metastasis,and the difference was statistically significant(P<0.05).3.The results of qRT-PCR and western blot showed that there were significant differences in the expression of PXMP4 in six colorectal cancer cells(P<0.05),among which HCT116 and SW620 were highly expressed,while RKO and SW480 were low expressed.4 The results of CCK-8 experiment showed that the growth rate of the overexpression group was significantly faster than that of the no-load group(P<0.05).The growth rate of cells in the interference group was significantly slower than that in the control group(P<0.05).5 The results of transwell invasion experiment showed that the number of cells in the overexpression group was significantly higher than that in the control group(P<0.05).However,the number of cells in the interference group was significantly less than that in the control group(P<0.05).6 The results of the scratch experiment showed that the migration rate of the overexpression group was significantly faster than that of the control group,while the migration rate of the interference group was significantly slower than that of the control group.The results of Transwell migration experiment showed that the number of cells in the overexpression group was significantly higher than that in the control group(P<0.05).However,the number of cells in the interference group was significantly less than that in the control group(P<0.05).Conclusions1.The expression of PXMP4 was significantly upregulated in colorectal cancer tissues,which was related to the degree of tumor differentiation and lymph node metastasis,but not to age,gender,or tumor size.2.PXMP4 promotes the proliferation,invasion,and migration of colorectal cancer cells.
