Study On The Therapeutic Synergistic Effect Of Chang Kang Fang Combined With Colon Targeted Probiotics On IBS-D Rats

Objective:By observing the effect of Chang Kang Fang combined with colon targeted probiotics on IBS-D rats,to explore whether Chang Kang Fang combined with colon targeted probiotics has synergistic effect on IBS-D rats,and to explore whether the mechanism is to regulate intestinal flora and improve the intestinal mucosal barrier function.Methods:The rat model was made by the method of senna leaf perfusion and binding stimulation.After the successful replication of the model,the rats were divided into 9 groups:high-dose Chang Kang Fang group,medium-dose Chang Kang Fang group,low-dose Chang Kang Fang group,colon targeted probiotics group,high-dose Chang Kang Fang combined with colon targeted probiotics group,medium-dose Chang Kang Fang combined with colon targeted probiotics group,low-dose Chang Kang Fang combined with colon targeted probiotics group,model control group and blank control group.There were 10 rats in each group.Two weeks after administration,the body weight growth rate,the number of defecation granules before and after 2 hours of modeling and 2 hours after administration were calculated,and the AWR score of rats was measured.At the same time,the change of intestinal flora was detected by high-throughput sequencing technology,and the expression of ZO-1 and TLR-4 were detected by immunohistochemistryResults:(1)Compared with the blank control group,the weight growth rate of rats in the model control group decreased,the number of defecation granules in the model group was significantly higher than that in the blank group after 2 hours of modeling(P<0.01),and AWR score increased under various pressure values(P<D.01).Therefore,it can be concluded that the model of IBS-D rats was successful,with a success rate of 80.95%.(2)After 2 hours of administration,the number of defecation granules in each intervention group was decreased in different degrees compared with the model group(P<0.05).Among them,the number of defecation particles in high-dose,medium-dose and low-dose Chang Kang Fang combined with colon targeted probiotics groups decreased more significantly(P<0.01).(3)After administration,the shannon index of model control group was significantly lower than that of blank control group,and the shannon index of each treatment group was significantly higher than that of model group.The ? diversity analysis showed that the intestinal flora diversity of IBS-D rats in each treatment group tended to the normal group,and the Chang Kang Fang combined with colon targeted probiotics group was more significant.After the treatment of Chang Kang Fang,colon targeted probiotics and Chang Kang Fang combined with colon targeted probiotics,the bacterial abundance of lachnospiraceae and ruminococcaceae in rats' intestines was significantly increased,and the bacterial abundance of prevotellaceae was decreased After the treatment with Chang Kang Fang combined with colon targeted probiotics,lactobacillus and clostridiurn were dominant in the intestine of rats.(4)After administration,the expression of ZO-1 protein in colon mucosa of model control group was significantly lower than that of blank control group,while the expression of ZO-1 protein in low-dose Chang Kang Fang group and colon targeted probiotic group was increased,but the degree of increase was limited(P<0.05).The expression of ZO-1 protein in high-dose,medium-dose and low-dose Chang Kang Fang combined with colon targeted probiotic group was relatively increased(P<D.01).Low-dose Chang Kang Fang combined with colon targeted probiotics group was particularly significant,with statistical significance.(5)The expression of TLR-4 in intestinal mucosa of rats in model control group was higher than that in blank control group.After administration,the expression of TLR-4 in intestinal mucosa of rats in each group was decreased to different degrees,but the effect of high-dose and medium-dose groups of Chang Kang Fang was less than that of model control group,with no statistical significance(P>0.05).The expression of TLR-4 in other groups was decreased with statistical significance(P<0.01).High-dose,medium-dose and low-dose Chang Kang Fang combined with colon targeted probiotics group decreased more significantly,while the most obvious was the low-dose Chang Kang Fang combined with colon targeted probiotics group.Conclusion:Chang Kang Fang combined with colon targeted probiotics can better improve the diarrhea symptoms of IBS-D rats.After the combination of the two drugs,the diversity of intestinal microflora in IBS-D rats tends to the blank control group,the expression of ZO-1 in intestinal mucosa increases,and the expression of TLR-4 decreases.It can be concluded that Chang Kang Fang combined with colon targeted probiotics has synergistic therapeutic effect on IBS-D rats,and its mechanism may be to regulate intestinal flora and improve intestinal mucosal barrier function.
