| Objective And Background:Nasopharyngeal carcinoma(NPC)is one of the most common malignant tumor diseases in China with a high degree of regional epidemic trend.Among the common cancers in otorhinolaryngology,the incidence of NPC is highest.Because the majority of NPC originates from occult sites such as the posterior parietal and lateral walls of the nasopharynx,it is highly invasive and metastasizes early.Most patients with nasopharyngeal carcinoma are in a state of local metastasis when seeking medical treatment.Thus,the search for diagnosis and treatment biomarkers of nasopharyngeal carcinoma provides a new treatment strategy for clinical practice,which will greatly improve the prognosis and quality of life of patients.In this study,based on whole transcriptome high-throughput sequencing experiments,we detected the expression of lncRNA CASC15 : 4 in NPC tissues and its correlation with the characteristics of a variety of clinicopathological factors by real-time PCR,in order to discover emerging tumor markers in the diagnosis and treatment of NPC and to provide individualized treatment for NPC patients.Methods:(1)The residual tissue samples of otolaryngologists undergoing endoscopic sinus biopsy were collected and 20 cases of nasopharyngeal non-cancer tissues and 30 cases of nasopharyngeal cancer tissues were obtained.All biopsies were confirmed as nasopharyngeal carcinoma tissue or chronic inflammatory tissue of the nasopharynx by the Department of Pathology of the First Affiliated Hospital of University of South China.All clinically relevant information was obtained from the information collection form completed by the examined patients.(2)Six cases of nasopharyngeal carcinoma or nasopharyngeal inflammation were divided into two groups.The expression profiles of long non-coding RNAs in the two groups were obtained by whole transcriptome high-throughput sequencing.(3)To verify the expression of lncRNA CASC15:4 in 30 NPC tissues and 20 benign nasopharyngeal tissues by using real-time PCR assay,and to analyze the correlation between lncRNA CASC15:4 and the characteristics of multiple clinicopathological factors in NPC patients.(4)IBM SPSS Statistics25.0 software was used to statistically analyze the experimental results.Results:(1)Results of whole transcriptome high-throughput sequencing of long non-coding RNAs in nasopharyngeal tissues(cancer and non-cancer): A total of 1392 long non-coding RNAs(lncRNAs)with statistically significant differential expression in nasopharyngeal tissues(cancer and non-cancer)(meeting the absolute value of fold difference greater than zero,and P value less than 0.05);a total of 1001 long non-coding RNAs with very significant differential expression(meeting the absolute value of fold difference greater than zero,and P value less than 0.001).Of the 1392 samples differentially expressed long non-coding RNA profiles,614 showed up-regulated expression trends and 778 showed low expression.(2)GO analysis of differentially expressed long non-coding RNAs revealed that the regulation of protein serine/threonine kinase activity was most enriched in biological processes;endosomal membrane was most enriched in cellular components;and cation channel activity was most enriched in molecular functions.(3)KEGG enrichment analysis of differentially expressed long non-coding RNAs revealed that 15 related pathways were associated with homeopathic regulatory coding genes of differentially expressed long non-coding RNAs,and the most significant pathway was the Intestinal immune network for IgA production pathway.(4)In the lncRNA differential expression profile obtained by whole transcriptome high-throughput sequencing,lncRNA CASC15:4 was up-regulated in NPC tissues;the expression content of lncRNA CASC15:4 was significantly higher in NPC tissues than in their expression in nasopharyngeal inflammatory tissues as verified by qRT-PCR experiments in human NPC tissues and nasopharyngeal inflammatory tissues,which was consistent with the sequencing results.(5)The expression level of lncRNA CASC15:4 was related to the clinical stage and differentiation of NPC patients,but not significantly correlated with the gender,age,smoking history and lymph node metastasis of patients.Conclusion:(1)The differential expression profile of long non-coding RNA in NPC tissues was obtained.(2)LncRNA-CASC15: 4 is abnormally highly expressed in nasopharyngeal carcinoma tissues,and it is closely related to the clinical stage and differentiation of patients with NPC.LncRNA-CASC15: 4 has a promoting role in the occurrence and development of nasopharyngeal carcinoma. |
