Effects Of CSRP1 On Proliferation,Apoptosis,Invasion And Migration Of Human Hepatocellular Carcinoma HepG2 Cells

Objective: To investigate the effect of overexpression of CSRP1(cysteine and glycine rich protein 1)gene on the proliferation,apoptosis,invasion and migration of HepG2 cells.Methods: HepG2 cells were cultured in vitro and transfected with pex-2-CSRP1 recombinant plasmid(overexpression group)or pex-2empty vector(empty plasmid group)respectively.Western blotting was used to detect the expression of csrp1 gene,MTT was used to detect the proliferation activity of HepG2 cells after 0h,24 h,48h and 72 h,and flow cytometry was used to detect the apoptosis of HepG2 cells.The invasiveness of HepG2 cells in each group was detected by Transwell invasiveness test,and the mobility of HepG2 cells in each group was detected by cell scratch test.Results: 1.Western blot results: 48 hours after transfection,the expression level of CSRP1 protein in HepG2 cells in the overexpression group increased significantly.The results of semi quantitative analysis showed that the relative expression of over expression group was(0.224± 0.0165),which was significantly higher than that of blank group(0.0723 ± 0.0138)and blank plasmid group(0.0730 ± 0.0143)(P < 0.05);2.MTT results: in three different time periods(24h,48 h,72h),the OD values of the overexpression group were(0.434 ± 0.025),(0.484 ±0.039),(0.619 ± 0.041),the blank group were(0.521 ± 0.408),(0.688 ±0.030),(0.912 ± 0.036),the empty plasmid group were(0.514 ± 0.024),(0.674 ± 0.028),(0.876 ± 0.027),respectively The OD value of the over expression group was significantly lower than that of the blank group and empty plasmid group(P < 0.05);3.The results of flow cytometry showed that the apoptosis rate of over expression group was(9.053 ± 0.304)%,higher than that of blank group(4.170 ± 0.212)%,empty plasmid group(5.097 ± 0.309)%,the difference was statistically significant(P < 0.05);4.The results of cell scratch test: in the three time periods(24h,48 h,72h),the cell migration distance of over expression group was(1.620 ±0.111)? m,(2.660 ± 0.707)? m,(3.640 ± 0.230)? m,lower than that of blank group(2.400 ± 0.158)? m,(3.720 ± 0.130)? m,(5.260 ± 0.230)?m and empty plasmid group(2.440 ± 0.219)? m,(3.820 ± 0.110)? m,(5.300 ± 0.245)? m,The difference was statistically significant(P <0.05).Conclusion: overexpression of CSRP1 gene can inhibit the proliferation,invasion and migration of HepG2 cells,and promote the apoptosis of HepG2 cells.
