Objective Allergic rhinitis is a multigene-regulated allergic disease,and its pathophysiological mechanism is complex.In this study,qRT-PCR was used to detect the concentration of long-chain non-coding RNA-RTL1 in nasal mucosa tissue,and to analyze its correlation with clinical indicators of allergic rhinitis.Methods 1.Blood samples were collected from 25 patients with allergic rhinitis(AR group)and 23 healthy people(control group),and serum-specific IL-4/10 levels were detected by ELISA;2.Collect nasal mucosa specimens from two groups of patients,and detect the expression of lnc-RTL1 in nasal mucosa tissues by fluorescence quantitative RT-PCR.Comparison of lnc-RTL1 concentration in nasal mucosa of AR patients and healthy people by statistical analysis;3.Subjective symptoms and quality of life scores for AR patients using VAS and RQLQ scales;4.Spearman correlation analysis was used to determine the relationship between the expression of lnc-RTL1 in AR nasal mucosa and AR-related clinical indicators(symptoms and quality of life scores,serum IL-4 and IL-10 levels)at the organizational level.Results 1.ELISA test showed that the median concentration of serum IL-4 in the AR group was 188.04 pg/mL and the control group was 148.048 pg/mL;the median concentration of serum IL-10 in the AR group was 291.586 pg/ml;the control group It was 431.608 pg/ml,and the difference was statistically significant(P<0.05).Serum IL-4 was higher in the AR group than in the control group,and serum IL-10 was lower in the AR group than in the control group.2.RT-PCR showed that the expression level of lnc-RTL1 was higher in the nasal mucosa of the allergic rhinitis group than in the control group.3.Correlation analysis between lnc-RTL1 and serum-specific IL-4/10 levels in AR nasal mucosa tissues using the Spearman Correlation Coefficient(PCC).There was no significant correlation between the expression of lnc-RTL1 in the AR group and the levels of serum IL-4 and IL-10,and the difference was not statistically significant(P>0.05).4.Correlation analysis of nasal mucosal lnc-RTL1 expression level with nasal symptoms and quality of life scores in patients with allergic rhinitis was performed by Spearman correlation.The results showed that the expression level of lnc-RTL1 in the nasal mucosa in the AR group was related to the subjective symptom score of AR patients,and the difference was statistically significant(P<0.001).Conclusion The expression of lnc-RTL1 in the nasal mucosa of allergic rhinitis and that of healthy people is different,but there is no significant correlation between the expression of lnc-RTL1 in the nasal mucosa of patients with allergic rhinitis and the serum specific IL-4/10 level,suggesting that lnc-RTL1 may not participate in the expression of serum IL-4/10 protein and may participate in the expression of other cytokine proteins,which may have gene regulation on the pathogenesis of allergic rhinitis.The expression of lnc-RTL1 in the nasal mucosa of patients with allergic rhinitis is inversely related to the subjective symptoms and quality of life scores,suggesting that lnc-RTL1 can be used as an evaluation index of the severity of allergic rhinitis.
