Objective:To establish a rat model of ventricular arrhythmia with xinyin deficiency syndrome,and observe the effect of Yangyin Dingji(YYDJ)decoction on ECG,left atrium and left ventricle electrical signals of rats with ventricular arrhythmia,so as to confirm the efficacy and possible mechanism of decoction on ventricular arrhythmiaMethods:Part one:60 Wistar rats were randomly divided into 6 groups according to the random number table method,with 10 rats in each group,which were the low,medium and high dose groups,positive control group,model group and blank group respectively.The model was built according to the idea of "combination diseases with syndromes".Except the blank group,the model of syndrome of xinyin deficiency was established by 4-week binding method combined with water platform sleep deprivation method in each group,and the model was evaluated from three aspects:general observation,behavioral evaluation and vital signs detection.The general conditions of the rats were observed every day,and the daily changes in diet,water and body weight of the rats were recorded before and after the modeling.The behavioral changes of rats were observed by opening experiment before and after modeling.The systolic blood pressure,heart rate,respiration and anal temperature were measured before and after modeling.After the success of the building,each group were given by gastric,low,medium and high dose group was given YYDJ particles of 0.41g/kg,0.81g/kg,1.62g/kg,positive control group given tartaric acid metoprolol 5.2mg/kg,blank control group and model control group given volume normal saline,groups are to fill the stomach,gastric volume are calculated according to weight,once a day.After 2 weeks of administration,BL-420F was used to monitor the electrocardiogram curve of ? lead and record the changes of electrocardiogram.Then,calcium chloride was injected into the tail vein to establish the model of ventricular arrhythmia syndrome of Yin deficiency of heart and compare the changes of electrocardiogram curve of rats before and after the injection.Part two:12 SD rats were randomly divided into 2 groups according to the random number table method,with 6 rats in each group,respectively as the control group and the observation group.The particles to be tested were directly dissolved in KH solution and prepared into 2%and 0.2%of the working fluid.The working fluid was then pushed into the heart for Langendorff perfusion.The left atrium and left ventricle were recorded by 128-channel matrix electrical labeling system to observe the effects of YYDJ decoction of different concentrations on the electrophysiology of the isolated heart of rats.Results:1.General observation:compared with the blank group,the average daily diet and body weight of the model group decreased after modeling,and the changes were significant in week 4(P<0.01).The average daily drinking water increased,and the change was significant in week 4(P<0.01).2.Behavioral evaluation:compared with the blank group,there was no statistical significance in the changes of horizontal movement,vertical movement and activity time within 5 minutes in the model group after modeling.However,it can be observed that the animal is hyperactive and agitated,and the activity path is mainly in the surrounding area,with less activity in the middle area,indicating that some model group rats can see anxiety3.Detection of vital signs:compared with the blank group,systolic blood pressure,heart rate,respiration and anal temperature were all higher in the model group after modeling(P<0.01).4.ECG:there was no significant difference in the incidence of ventricular premature beats(VP),ventricular tachycardia(VT)and ventricular fibrillation(VF)in each group.Compared with the model group,the time of VP in rats in the middle and high dose YYDJ decoction showed a delayed trend,and the time of VP in the middle dose YYDJ decoction was 35.7±10.4(s)significantly delayed(P<0.05).The time of VP in the high-dose YYDJ decoction group was 37.5±5.6(s)(P<0.05).Compared with the model group,the time of cardiac arrest was delayed in each dose group(P<0.01)5.Electrical signal detection:YYDJ decoction with 0.2%and 2%concentration could slow the heart rate of rats,decreasing from 302.33±19.98 BPM to 282.50±27.46 BPM and 228.00±13.87 BPM,respectively.At the same time,YYDJ decoction with 2%concentration had a strong inhibitory effect on the conduction of atrium and ventricle,which reduced the conduction speeds of atrium and ventricle to 46.09±2.51%and 57.90±12.07%of the control group,respectively.The 2%concentration of YYDJ decoction can slightly increase the atrioventricular delay timeConclusion:1.In this study,the model of ventricular arrhythmia with Yin deficiency could be successfully constructed by binding method and water platform sleep deprivation method and injecting calcium chloride into tail vein.2.YYDJ decoction has the effect of resisting the ventricular arrhythmia,presenting obvious delayed ventricular morning and ventricular tachycardia at medium and high dose,and shortening the duration3.The administration of YYDJ decoction at 0.2%and 2%concentration could slow the heart rate of rats,the administration of 2%concentration had a strong effect on the conduction of atria and ventricles,and the administration of 2%concentration could slightly increase the atrioventricular delay time.
