Objective: This subject aimed to use electroacupuncture treatment with different acupoints after constructing a spinal cord injury model.By observing the behavioral score,Basso,Beattie and Bresnahan locomotor rating scale(BBB score)of rats after spinal cord injury,pathological changes of the spinal cord,glial-derived acidic protein(GFAP)after spinal cord injury express the characteristics of changes,evaluate the efficacy of electroacupuncture compatibility with different points,and whether there is a difference in efficacy between different acupoint combinations,so as to find a more reasonable electroacupuncture point compatibility program to provide experimental data and theoretical basis for clinical acupuncture treatment.Methods: In this experiment,100 SD rats,male and female,were divided into sham group,model group,and an electroacupuncture treatment group(EA1,EA2,and EA3groups)according to random number method.Each group had 20 rats,and each group was based on different times.The window was divided into 4 subgroups(3d,7d,12 d,and 18 d postoperatively),5 in each subgroup.Using the improved Allen's method of the research group to build a spinal cord injury model.The acupuncture points in the electroacupuncture treatment group were selected as follows: Dazhui,Mingmen,Jiaji,Zusanli,Sanyinjiao in EA1 group,Dazhui,Mingmen,Jiaji,Shuigou,Yanglingquan in EA2 group,Dazhui,Mingmen,Jiaji,Shenshu,Ciliao in EA3 group.Electroacupuncture intervention was performed 1 day after surgery,once a day for 30 min each time,a course of 5 days,and a rest period of 2 days during the course of treatment.BBB scoring was used to evaluate changes in hindlimb motor function before and after modelling.Observing the tissue pathological structure,Nissl body and GFAP after spinal cord injury with HE staining,the Nissl staining,immunohistochemical techniques,and the difference in curative effect between the EA group and the model group was compared.Summarize and analyze the difference in efficacy between the treatment group and the model group.Results: 1.The results of BBB score:At different time points after spinal cord injury,the model group's hindlimb motor function score was significantly reduced.After 3d and7 d treatment,there was no significant difference in the scores of hindlimb motor function of each electroacupuncture group compared with the model group.After 12 d treatment,the scores of hindlimb motor function of the EA1 and EA2 groups increased compared with the model group,with statistics Significance(P ? 0.01),but there was no statistically significant difference between the EA3 group and the model group.After 18 days of treatment,the limb motor function scores of the three groups of electroacupuncture groups were significantly increased,which was significantly different from the model group(P?0.01).2.HE staining:The sham operation group had a complete spinal cord structure and clear neuronal morphology.In the model group,3d after spinal cord injury,extensive hemorrhage of the spinal cord tissue was seen,and the normal structure of the spinal cord was destroyed;on 7d and 12 d,the spinal cord tissue was severely damaged,and necrotic cells,glial cells increased,and vacuoles were formed;on 18 d,the spinal cord tissue was loose and cells Shrink and solidify.After electroacupuncture treatment in different groups,there was no significant change in spinal cord tissue at 3d,neurons were repaired at 7d and12 d,and cell necrosis was reduced.The effect was better at 18 d after treatment.3.Nissl staining:The structure of spinal cord and neurons in the sham-operated group was complete and clear,and full Nissl bodies were visible.The spinal cord gray matter and white matter structure were destroyed 3d and 7d after spinal cord injury.Nissl bodies of neurons became fewer,smaller or disappeared.Neurons contracted,nucleus cracked,and even dissolved and liquefied.Large vacuolar changes were seen around the neurons at 12 d and 18 d.There was no obvious recovery of Nissl bodies of neurons on 3d,7d and 12 d after electroacupuncture treatment in different groups.The number and morphology of Nissl bodies of neurons were repaired on 18 d after treatment.4.Immunohistochemical staining:GFAP-positive cells were located in the cytoplasm and developed yellow-brown color.When the spinal cord tissue was not injured,the expression of GFAP showed that the number of astrocytes was small,the cell body wassmall,and there were few protrusions;3 days after the spinal cord injury,no obvious GFAP positive cells were seen;7 days and 12 days after the injury,the number of GFAP positive cells gradually increased,but the cell body Slender shape;18 days after injury,the number of GFAP-positive cells increased significantly,the cell body increased,and the proliferation of cells at the edge of the lesion was more obvious.After electroacupuncture treatment in different groups,the number of GFAP-positive cells did not change significantly at 3d,but at 7d and 12 d,the cell body of GFAP-positive cells was obviously hypertrophic,and at the same time,the number of protrusions increased,and the edge of the lesion proliferated significantly.Conclusion: The results of BBB scoring showed that the three groups of different electroacupuncture groups could improve the motor function of the limbs after spinal cord injury in rats.After 12 days of treatment,the motor function scores of the three groups of treatment groups had increased significantly,and the effect was more significant after 18 days of treatment.The efficacy of the EA1 and EA2 groups was better than that of the EA3 group.The HE staining and Nissl staining showed that three groups of different electroacupuncture groups can significantly improve the tissue structure and pathological changes of Nissl body after spinal cord injury in rats,protect neurons and reduce the number of cell damage.The effect is better after 18 days of treatment.However,there was no significant difference in efficacy between the three electroacupuncture treatment groups.Immunohistochemical staining showed that after treatment with three different electroacupuncture groups,the number of GFAP-positive cells did not change significantly at 3 days,but the cell bodies of GFAP-positive cells at 7 and 12 days were significantly enlarged,and at the same time,the number of protrusions increased and the proliferation of cells at the edge of the lesion was obvious And it showed an increasing trend,reaching a peak at 12 d,the number of GFAP positive cells decreased at 18 d,the cell body still increased,but the cell body protrusion decreased.The positive area ratio of GFAP in the three treatment groups was significantly different from that in the model group,but therewas no significant difference in efficacy between the three treatment groups.In summary,after electroacupuncture treatment of different acupoints in the treatment of spinal cord injury,the hindlimb motor function of rats was significantly improved,and the structure of spinal cord and Nissl body were repaired,and it was beneficial to the recovery of spinal cord injury by regulating the expression of GFAP.
