| Objective:In this study,Mongolian medicine sugmule-3 Decoction(SD-3)was used to improve isoproterenol(ISO)-induced cardiac hypertrophy in rats by mediating the expression of calumenin protein.The effect of calumenin protein and calcium overload on the treatment of myocardial hypertrophy in rats was studied in vivo and in vitro.Methods:Wistar male rats and H9c2 cell lines were used for in vitro and in vivo experiments.The model of myocardial hypertrophy and cardiomyocyte hypertrophy was established by ISO.Three different concentrations of sugmule-3 Decoction and metoprolol were used.At the animal level,echocardiography and ECG were used to observe the cardiac function indexes of rats in each group,Masson staining was used to observe the pathological changes of rat heart,and TUNEL staining and AO/EB fluorescence staining were used to detect the apoptosis of rat cardiac myocytes at the cell level.Western blot and RT-PCR were used to detect cardiac hypertrophy related indexes,calumenin protein,calcium channel and potassium channel related indexes,endoplasmic reticulum stress channel related proteins and genes in vitro and in vivo.Result:In vivo,SD-3 was found to significantly improve cardiac hypertrophy and arrhythmia in rats.It was preliminarily confirmed that SD-3 could further affect L-type calcium current channel,Na+/Ca+exchange current channel,inward rectifier potassium current channel and instantaneous outward potassium current channel by up regulating the expression of calumenin protein;delayed rectifier potassium current channel can alleviate ER stress and improve myocardial hypertrophy induced by ISO.In vitro experiments confirmed that SD-3 could further affect L-type calcium current channel,Na+/Ca+exchange current channel,delayed rectifier potassium current channel and transient outward potassium current channel by up regulating the expression of calumenin protein,and ultimately improve the apoptosis of rat cardiomyocytes.Conclusion:The results of animal experiments show that Mongolian medicine sugmule-3 decoction can significantly improve the phenomenon of myocardial hypertrophy in rats,and its effect may be through mediating the expression of calumenine protein to affect calcium overload to further alleviate endoplasmic reticulum stress and ultimately affect the occurrence of apoptosis.The results of cell experiments showed that medicine sugmule-3 decoction could significantly improve the hypertrophic phenomenon of rat cardiomyocytes,and its effect might be through mediating the expression of calumenine protein to affect calcium overload to further alleviate endoplasmic reticulum stress and ultimately affect the occurrence of apoptosis. |
