| Objective(s):To observe the effect of Suppressor of cytokine signaling3(SOCS3)on the migration and invasion of human gallbladder carcinoma cells(GBC-SD).To investigate the effect of SOCS3 on casein kinase 2/signal transduction and transcriptional activator 3(JAK2/STAT3)signal pathway,so as to investigate the mechanism.Methods:1.The three siRNA gene fragments that interfered SOCS3 site synthesized by Shanghai gima gene company were RAN oligo socs3-homo-615,778 and 991,respectively.After siRNA transfection with Lipofectamine3000,the expression of SOCS3 after interference was verified by Western Blot detection and cell slide immunohistochemistry,and the siRNA fragment with the best interference effect was selected as the interference fragment in the experimental group.2.Cell scratch assay and Transwell assay were used to detect the effect of SOCS3 inhibition on the migration and invasion ability of gallbladder cancer cells.3.With selected effective siRNA interference fragment transfection to gallbladder cells(GBC-SD)as experimental group,transfection the corresponding negative control sequence of siRNA group,as the negative control group did not make any sequence transfection gallbladder cells as blank control group,by using Western Blot detection JAK2/STAT3 signaling pathway of protein and total JAK2,total STAT3 phosphorylation casein kinase 2(p-JAK2),signal transduction and transcriptional activation factor 3(p-STAT3)expression of the situation,To investigate the effect of SOCS3 down-regulation on JAK2/STAT3 signaling pathway in gallbladder carcinoma cells(GBC-SD).Results:1.Three siRNA fragments of gallbladder carcinoma cells(GBC-SD)were transfected with Lipofectamine3000 liposome transfection reagent,and the expression of SOCS3 was detected by Western Blot.It was found that RAN oligo socs3-homo-615 significantly interfered with the down-regulation of SOCS3 expression.Immunohistochemistry experiments of cell slide confirmed the good interference effect of the fragment.2.By cell scratch test,it was found that down-regulating the expression of SOCS3 in human gallbladder cancer cells gbc-sd inhibited the migration ability of gallbladder cancer cells.Transwell assay showed that down-regulation of SOCS3 expression in cells could reduce the invasion ability of cells.3.Interference down-regulated the expression of SOCS3 in GBC-SD of human gallbladder cancer cells,and JAK2/STAT3 signaling pathway protein decreased.Conclusion(s):1.Interference down-regulated the expression of SOCS3 in gallbladder cancer cells(GBC-SD),and the migration and invasion ability of gallbladder cancer cells decreased.SOCS3 may play a role in promoting tumor proliferation,migration and invasion in gallbladder cancer.2.In gallbladder cancer cells(GBC-SD),SOCS3 may affect the invasion and migration of gallbladder cancer cells through the JAK2/STAT3 signaling pathway. |
