Protective Effect Of PDGF-AA On Neonatal Islet Cells In Vitro And Its Mechanism

Objectives:The protective effect of PDGF-AA on neonatal islet cells in vitro was studied,and the possible molecular mechanism was analyzed.Methods:1.DTZ staining and AO/PI staining were used to evaluate the protective effect of PDGF-AA on the morphology and activity of islet cells in vitro.2.MTS was used to detect the protective effect of PDGF-AA on the proliferation capacity of islet cells in vitro.3.The protective effect of PDGF-AA on the function of islet cells in vitro was detected by the experiment of glucose stimulating insulin secretion.4.Western Blot was used to detect Bax,Bcl-2 in islet cells treated with PDGF-AA.changes in Ezh2 protein expression.Results:1.The results of DTZ staining and AO/PI staining showed that the islet cells treated with PDGF-AA had better morphology and activity in vitro than the control group(P<0.05).2.MTS results showed that islet cells treated with PDGF-AA had better proliferation capacity in vitro than those in the control group(P<0.05).3.Glucose stimulated insulin secretion experiment results showed that the pancreatic islet cells treated with PDGF-AA had stronger insulin secretion capacity than the control group cultured in vitro in response to glucose stimulation(P<0.05).4.Western Blot results showed that islet cells treated with PDGF-AA could increase the expression of Ezh2 protein in islet cells compared with the control group(P<0.05),but had no effect on the expression of Bax protein(P>0.05),and no effect on the expression of Bcl-2 protein(P>0.05).Conclusions:1.Islet cells treated with PDGF-AA can improve the morphological activity of islet cells in vitro.It can improve the proliferation of islet cells in vitro and the function of islet cells in vitro.PDGF-AA can protect islet cells to a certain extent in vitro.2.Islet cells treated with PDGF-AA can increase the expression of Ezh2 in islet cells,and play a protective role in vitro by increasing the expression of Ezh2.However,there was no effect on the expression of Bax and bcl-2 in islet cells.
