Objective:The homing effect of fat source mesenchymal stem cells on hematopoietic stem cells after transplantation and the preliminary exploration of tissue repair support.Methods:ADSCs,BMSCs and RF were obtained from C57BL/6 male mice under strict aseptic conditions to prepare Fibroblast cells.After culture to P3,cell morphology was observed,and adipogenic and osteogenic differentiation of ADSCs was conducted.Oil red O and Alizarin red staining was used to identify the multidirectional differentiation function of ADSCs.LSK cells were labeled with DID~+,and ADSCs and RF cells were labeled with CSFE~+.Rat BABL/C was designed and grouped by means of myeloablative pretreatment:(1)Blank control;(2)1500 DID+LSK cells;(3)1500 DID+LSK cells and 1x106 CFSE+RF;(4)1500DID~+LSK 1x10~6CFSE~+ADSCs.The mice were injected in groups by tail vein,and 24 hours later,the mice were sacrificed and bone,liver,spleen,lung,intestine,heart and kidney were collected for wax block embedding,and wax block pathological sections were made and stained with HE.Results:The cells obtained from C57BL/6 adipose issue are MSC cells with multidirectional differentiation.Pathological sections of BABL/C mice treated with ADSCs+LSK infusion showed that ADSCs could promote homing and hematopoietic recovery of LSK cells.Fluorescently labeled DID+LSK cells,CSFE+ADSC,and CSFE+RF cells were more distributed in bone after infusion by tail vein in mice,and no obvious concentration was observed in other organs.Conclusions:ADSCs promoted the homing effect of LSK in vivo.At present,it may be related to immune regulation,tissue repair,endocrine and paracrine functions of ADSCs.HE pathological sections and cell fluorescence distribution analysis of the sacrificed mice showed that ADSCs and LSK were mostly distributed in bone.
