MiR-214-3p Inhibits The Osteogenic Differentiation Of Dental Follicle Cells Through Beta-catenin/TCF-4 Signaling Pathway In Vitro

Objective: To investigate the effect of microRNA-214-3p(microRNA-214-3p)on the osteogenic differentiation of dental follicle cells(DFCs)through beta-catenin/TCF-4 signaling pathway.Methods: Dental follicle cells were isolated,cultured an d purified by Bidirectional differential method.The expression of microRNA-214 in DFCSs was up-regulated and down-regulated by transfecting miR-214 mimics and miR-214 inhibitors into DFCs.After 7 days of osteogenesis induction,the expressi on of microRNA-214 and related osteogenesis genes,as well as the changes of bet a-catenin/TCF-4 pathway related proteins,were confirmed by qRT-PCR,alizarin red staining and Western-blot.Results: RT-q PCR results showed that:(1)The expres sion of miR-214 decreased after osteogenesis induction of DFCs.(2)miR-214 mim ics and inhibitor were transfected into DFCs by Lipo3000,which could up-regulate and down-regulate the expression of miR-214 in DFCs.(3)The up-regulation of microRNA-214 can down-regulate the osteogenesis-related genes ALP(P<0.05),OS N(P>0.05),RUNX-2(P>0.05)and miR-214 can up-regulate the osteogenesis-relate d genes ALP,OSN and RUNX-2(P<0.05).Western blot assay showed that up-reg ulation of microRNA-214 could down-regulate the expression of beta-catenin/TCF-4signaling pathway-related proteins and inhibit osteogenic differentiation.If microR NA-214 was down-regulated,the result was the opposite.Alizarin red staining ima ges showed that the mineralized nodules of DFCs transfected with miR-214 mimics and miR-214 inhibitor were significantly different after osteogenesis induction.Co nclusion:(1)The expression of microRNA-214-3p in DFCs decreased after osteogenesis induction.(2)miR-214-3p may regulate the osteogenic differentiation of DFC s through the beta-catenin/TCF-4 signaling pathway.
