Study On The Mechanism Of Sevoflurane Preconditioning To Optimize Glycolysis Of Cardiomyocytes To Alleviate Hypoxia/Reoxygenation Injury

Abstract/Summary:
Objective:To investigate whether sevoflurane preconditioning can regulate the anaerobic glycolysis of cardiomyocytes H9c2 by up regulating signal path HIF-1/PDK-1 and ultimately reduce the hypoxia / reoxygenation injury of cardiomyocytes.Methods: After cultured in low glucose medium for 48 hours,H9C2 cardiomyocytes were randomly divided into four groups:(1)control group(CON);(2)hypoxia / reoxygenation group(H/R);(3)sevoflurane preconditioning group(SPC);(4)sevoflurane preconditioning group + HIF-1a inhibitor group(S + YC-1);Establish hypoxia / reoxygenation model,three hours after reoxygenation,LDH、HK、PFK-1 and PK were detected;flow cytometry was used to detect apoptosis rate and membrane potential;ATP energy was detected;Western blot was used to detect HIF-1a,PDK-1,GAPDH and apoptosis-related proteins.Results: Compared with H/R group,the contents of key enzymes(HK,PFK-1,PK)of glycolysis、the amount of ATP、the mitochondrial membrane potential and the content HIF-1a、PDK-1、Bcl-2 in SPC group increased significantly,but the content of Bax protein and the proportion of apoptosis were decreased(P<0.05);Compared with SPC group,the content of glycolysis key enzymes(HK,PFK-1,PK),ATP production,mitochondrial membrane potential,HIF-1a,PDK-1,Bcl-2 protein decreased significantly,while the proportion of apoptosis and Bax content of apoptosis-related protein increased significantly(P<0.05).Conclusions: Sevoflurane preconditioning optimizes glycolytic energy metabolism of myocardial cells by activating HIF-1a / PDK-1 signaling pathway,and reduces hypoxia / reoxygenation injury of H9C2 cardiomyocytes.
